重组生长激素改善小鼠子宫内膜损伤疗效分析 被引量:1

Study of growth hormone on repair and receptivity of damaged endometrium in mice

摘要 目的研究重组生长激素(GH)能否修复小鼠受损的子宫内膜及其对子宫内膜容受性的影响。方法通过宫腔内灌注95%乙醇建立受损的小鼠子宫内膜模型,重组生长激素采用皮下注射及宫腔灌注两种形式,经过不同方式治疗的小鼠作为实验组,建模后宫腔灌注生理盐水及未经任何处理的小鼠作为对照组。分别治疗2个动情周期后检测子宫内膜并进行生育测试。HE染色检测子宫内膜厚度、腺体及血管数目。采用免疫组织化学法观察雌激素受体(ER1)、孕激素受体(PR)、整合素ανβ3及骨桥蛋白在两组子宫内膜组织中的表达情况评价子宫内膜容受性。结果构建小鼠的子宫内膜损伤模型,各组小鼠均有不同程度的盆腹腔粘连及子宫肿胀,以宫腔灌注生理盐水组最严重。重组生长激素两种不同方式的治疗后,小鼠受损的子宫内膜组织结构都有不同程度的恢复,子宫内膜厚度、腺体数目及血管分布均高于宫腔灌注生理盐水组,但是两种治疗方式在改善子宫厚度、改善子宫内膜容受性差异无统计学意义(P>0.05)。结论重组生长激素可以修复受损的子宫内膜,提高子宫内膜容受性。 Objective To investigate whether growth hormone can repair damaged endometrium in mouse and to estimate its effect on endometrial receptivity.Methods The mouse endometrial damage model was established by intrauterine perfusion of 95%ethanol.Growth hormone was used by subcutaneous injection and intrauterine perfusion.Mice treated in different therapeutic ways of growth hormone were selected as the experimental group,while mice perfused with normal saline and normal mouse without damage were selected as the control group.The endometrium and fertility testing were examined after two estrous cycles.Endometrial thickness,the number of glands and blood vessel were examined to assess the repair of damaged endometrium by HE staining.The expression of estrogen receptor1,progesterone receptor,integrinανβ3 and osteopontin was used to evaluated the endometrial receptivity.Results Pelvic and peritoneal adhesions and endometrial swelling were happened in model mice of each group,among which the mice intrauterine perfusion with normal saline were most serious.Compared to control group which were perfused with normal saline,the endometrial structure recovered after the treatment with growth hormone.Endometrial thickness,the number of glands and blood vessel were increased significantly.But there were no significant difference in two therapeutic groups(P>0.05).Conclusion Growth hormone can repair damaged endometrium and improve the endometrial receptivity.
