摘要
【目的】研究胰岛素样因子3(Insulin-like factor 3,INSL3)对滩羊睾丸发育及生殖机能调节的影响。【方法】采用睾丸摘除手术收集初生及35日龄滩羊睾丸,应用特殊染色,免疫组织化学及免疫荧光技术方法结合形态计量学统计软件,比较初生与35日龄滩羊睾丸的组织化学特点,分析滩羊睾丸组织化学特点及发育规律。【结果】相较于初生滩羊,35日龄滩羊睾丸组织发育,AB染色在生精小管基膜阳性反应增强;生殖母细胞PAS染色阳性反应明显,间质组织胶原纤维增加。免疫组化及免疫荧光结果表明,35日龄滩羊睾丸组织中INSL3在生殖母细胞、Sertoli细胞及管周肌样细胞中表达较初生滩羊有所减弱;在Leydig细胞中表达下降。【结论】滩羊初生至35日龄睾丸发育过程中,酸性糖类物质代谢优于其他糖类成分,网状纤维生成早于胶原纤维。INSL3对睾丸生精细胞无明显调节作用;其表达降低与Leydig细胞中FLCs细胞群逐渐减少相一致。本研究为INSL3对Leydig细胞分泌功能调控机制的研究提供了参考。
【Objective】This study aimed to research the histochemical characteristics and effect of Insulin-like factor 3(INSL3)on the development of Tan sheep testis.【Method】The testicular histochemical characteristics were compared by special staining,immunohistochemistry,immunofluorescence and morphometric statistical software between the newborn and 35-day-old Tan sheep.【Result】Compared with the newborn Tan sheep,the testicular tissue of the 35-day-old Tan sheep developed obviously.The positive reaction of AB staining on the basal lamina of seminiferous tubule and the PAS staining of gonocytes were increased prominently,and there were abundant collagen fibers in interstitial tissue.The INSL3 expressed decreased in gonocytes,Sertoli and peritubular myoid cells of 35-day-old Tan sheep testis compared with those at newborn from the results of immunohistochemistry combining with immunofluorescence.The results also showed that the expression in Leydig cells decreased with age.【Conclusion】Overall,the metabolism of acid glycogen was superior to other carbohydrate components during the testis development of Tan sheep from birth to 35 days of age,and the formation of reticular fibers was earlier than that of collagen fibers.The decrease of INSL3 was consistent with the gradual decrease of FLCs numbers.This study could provide a reference for the study on the regulation mechanism of INSL3 in Leydig cell secretory function.
