摘要
优化超声辅助提取节茎石仙桃多糖的工艺参数并探究其抗氧化活性。在单因素试验基础上,采用Box-Behnken设计试验方法,探讨料液比、浸取温度、超声时间3个因素及其交互作用对节茎石仙桃多糖提取率的影响,构建节茎石仙桃多糖提取率与提取过程中各因素的二次多元回归模型。以抗坏血酸为对照,考察节茎石仙桃多糖对·OH、O^(2-)、ABTS·+和DPPH·的清除活性。结果表明,超声辅助水提醇沉节茎石仙桃多糖的最佳工艺条件为:料液比1∶30(g/mL),浸取温度78℃(1 h),超声时间38 min(80℃),在此提取条件下节茎石仙桃多糖的提取率为11.31%,与预测值仅相差0.11%,验证了该模型的有效性。节茎石仙桃多糖对·OH、O^(2-)、ABTS·^(+)和DPPH·4种氧化因子的综合清除活性较强,平均清除率达到(56.00±0.26)%以上,且多糖浓度与清除活性呈正相关。说明该工艺对石仙桃属植物多糖纯品的制备和开发利用均具有良好的应用价值。
In order to optimize the ultrasonic assisted extraction process parameters and explore the antioxidant activity of polysaccharide from Pholidota articulata Lindl.,on the basis of single factor experiment,Box-Behnken design experimental method was used to explore the effects of three factors such as material-liquid ratio,leaching temperature and ultrasonic time and their interaction on the extraction rate of polysaccharide from Pholidota articulata Lindl..A quadratic multiple regression model between the extraction rate of polysaccharide from Pholidota articulata Lindl.and various factors in the extraction process was constructed.Using ascorbic acid as control,the scavenging antioxidant of polysaccharide from Pholidota articulata Lindl.on·OH,O^(2-),ABTS·^(+)and DPPH·were investigated.The results showed that the optimum technological conditions of ultrasonic assisted water extraction and alcohol precipitation were as follows:material-liquid ratio of 1∶30(g∶mL),extraction temperature 78℃(1 h)and ultrasonic time 38 min(80℃).Under these extraction conditions,the extraction rate was 11.31%,which was only 0.11%different from the predicted value.The polysaccharide had an average scavenging rate of more than(56.00±0.26)%on the above four oxidation factors,and the polysaccharide concentration was positively correlated with the scavenging capacity.Although it was still insufficient compared with VC,it had obvious antioxidant activity.It shows that the process has good application value for the preparation,development and utilization of pure polysaccharides from Pholidota plants.
