摘要
目的优化中药材牛膝的薄层鉴别方法,为其质量控制提供方法和参考。方法对2020版药典中的牛膝薄层色谱法进行优化,以乙醇为提取溶剂,齐墩果酸为对照品,牛膝为对照药材,甲苯-乙酸乙酯-甲酸(20∶5∶2.5)为展开剂,在硅胶G薄层板上展开,喷以5%香草醛硫酸溶液,105℃加热至斑点显色清晰。结果供试品溶液与对照药材、对照品相应位置上显示相同颜色斑点,且斑点清晰,Rf值适当。结论所建立的牛膝薄层鉴别方法操作简单、耐用性佳、重现性好,可替代原标准用于牛膝的鉴别。对比相同等级药材齐墩果酸斑点亮度,结果显示同等级不同产地牛膝药材质量:四川产地>内蒙产地>河南产地>河北产地。
OBJECTIVE To improve a thin layer chromatography identification method of Chinese herbal medicine Achyranthes Bidentatae,and to provide method and basis for quality control standard.METHODS Improvement of Achyranthes bidentata thin layer chromatography method in the 2020 edition pharmacopeia:Ethanol was used as the extraction solvent;Oleanolic acid was used as controls;Achyranthes bidentatae was used as control herbs;and butyl methylbenzene-ethyl acetate-formic acid was used as the developing agent(20∶5∶2.5).The expansion was carried out on the silica gel-G plate.Vanillin sulfuric acid solution(5%)as the color developing reagent,heated at 105℃until the spots were clear.RESULTS The test solution showed clear spots of the same color at the corresponding positions with the contrast material and the reference material,and the Rf value was appropriate.CONCLUSION The established thin layer identification method of Achyranthes bidentata has simple operation,good durability,good reproducibility,and can replace the original standard for the identification of Achyranthes bidentata.Comparing the brightness of oleanolic acid spots of the same grade of medicinal materials,the results showed that the quality of the medicinal materials of the same grade and different origins:Sichuan origin>Inner Mongolia origin>Henan origin>Hebei origin.
