摘要
目的建立药材艾不同部位中4种活性成分的高效液相色谱(HPLC)含量测定方法。方法采用HPLC测定艾叶和茎中绿原酸、芦丁、槲皮素、山奈酚含量,色谱柱为Agilent Extend C_(18)柱(4.6 mm×250 mm,5μm);流动相为乙腈-0.1%磷酸水溶液,梯度洗脱;检测波长为360 nm;柱温为30℃;流速为1.0 ml/min。结果绿原酸、芦丁、槲皮素、山奈酚分别在7.826~782.6μg/ml,0.842~84.2μg/ml,1.685~168.5μg/ml,2.654~265.4μg/ml范围内与峰面积线性关系良好(R^(2)分别为0.9998,0.9995,0.9997,0.9998),精密度、稳定性和重复性良好,平均加样回收率分别为98.92%,97.58%,101.25%,98.02%,RSD分别为1.89%,2.04%,1.69%,2.27%。艾叶和茎中4个成分的含量存在显著差异,其中绿原酸在叶和茎中含量最高,芦丁在叶和茎含量最低。结论本研究所建立的方法高效简便、专属性强,可用于艾叶和茎中绿原酸、芦丁、槲皮素、山奈酚的含量测定。
Objective To develop a high performance liquid chromatography(HPLC)method for the simultaneous determination of four components in different parts of Artemisia argyi Levl.et Vant.Methods The contents of chlorogenic acid,rutin,quercetin and kaempferol were determined by HPLC on Agilent Extend C_(18)column(4.6 mm×250 mm,5μm),with acetonitrile-0.1%phosphoric acid as the mobile phase.The detection wavelength was 360 nm.The column temperature was 30℃,and the flow rate was 1.0 ml/min.Results Chlorogenic acid,rutin,quercetin and kaempferol showed a good linear relationship with peak area in the concentration range of 7.826-782.6μg/ml,0.842-84.2μg/ml,1.685-168.5μg/ml,2.654-265.4μg/ml,respectively,and R^(2)were 0.9998,0.9995,0.9997,0.9998,respectively.The precision,stability and repeatability of the method were good.The average recoveries were 98.92%,97.58%,101.25%and 98.02%,respectively,and the RSDs were 1.89%,2.04%,1.69%and 2.27%,respectively.There was significant difference in the contents of the four components in the leaves and stems of Artemisia argyi Levl.et Vant.The content of chlorogenic acid was the highest in leaves and stems,and the content of rutin was the lowest in leaves and stems.Conclusion The method established in this study is efficient,simple and specific,and can be used for the determination of chlorogenic acid,rutin,quercetin and kaempferol in Artemisia argyi Levl.et Vant leaves and stems.
