qPCR和胶体金试纸条方法检测猫疱疹病毒感染的比较

Comparison of qPCR and Colloidal Gold Test Strip for Detection of Feline Herpesvirus Infection

本研究比较猫疱疹病毒(FHV-1)2种检测方法荧光探针qPCR和胶体金试纸在检测FHV-1时各自的优劣,并以PCR扩增FHV-1特异基因Glycoprotein B序列并测序的方法作为参比方法。采集107份疑似FHV-1感染样本,分别用3种方法进行检测,qPCR方法采用的是冻干型PCR-荧光探针法,胶体金试纸方法采用的是市场成熟产品,参比方法测序结果与GeneBank数据进行比对,确认样本是否为FHV-1。qPCR检出89例FHV-1阳性,18例FHV-1阴性,胶体金试纸条检出51例FHV-1阳性,56例FHV-1阴性,参比方法检出89例阳性,且基因序列比对确认为FHV-1。通过对比实验结果的分析,qPCR方法灵敏度为100%,胶体金试纸条方法灵敏度为57.3%,qPCR检测灵敏度明显优于胶体金试纸。

In this study,the advantages and disadvantages of two detection methods of feline herpesvirus(FHV-1),qPCR and colloidal gold test paper,were compared in the detection of FHV-1.The method of PCR amplification of FHV-1 specific gene Glycoprotein B sequence and sequencing was used as a reference method.A total of 107 suspected FHV-1 infected samples were collected and detected by three methods.The qPCR method used the freeze-dried PCR-fluorescent probe method,and the colloidal gold test paper method used the market mature product.The sequencing results of the reference method were compared with the GeneBank data to confirm whether the sample was FHV-1.89 cases of FHV-1 positive and 18 cases of FHV-1 negative were detected by qPCR,51 cases of FHV-1 positive and 56 cases of FHV-1 negative were detected by colloidal gold test strip,89cases of positive were detected by reference method,and the gene sequence alignment was confirmed as FHV-1.By comparing the experimental results,the sensitivity of qPCR method was 100 %,and the sensitivity of colloidal gold test strip method was 57.3 %.The sensitivity of qPCR detection was significantly better than that of colloidal gold test strip.
