辽宁乡土杨树重测序遗传进化全基因组关联分析

摘要 为揭示辽宁省乡土杨树遗传进化和多样性,该文对辽宁省主要乡土杨树14个样品进行重测序,开展了遗传进化全基因组关联分析和数据评估。本次分析数据量为75.57 Gbp的Clean Data,Q30达到93.77%。测序数据质量质控以及与参考基因组对比统计结果显示:与参考基因组平均比对率为96.72%,基因组覆盖深度为84.92%(至少一个碱基覆盖),平均覆盖深度为10X。通过样品的SNP和InDel的检测和注释,进行了遗传进化和遗传多样性分析,并对乡土杨树群体进行了评价。从系统发育变异检测结果看,白杨派发育早,与黑杨派、青杨派在系统发育中亲缘关系较远,青杨派种间有基因交流现象。该项研究为乡土杨树资源保护和利用提供参考。 In order to reveal the genetic evolution and diversity of native poplar in Liaoning Province,Genome-wide association study(GWAS)was used to analyze and evaluate genetic evolution on 14 samples of major native poplar in Liaoning Province.75.57Gbp clean data of genome were obtained by GWAS,and the Q30 reached 93.77%.The statistical results of sequencing data quality control and comparison with reference genome showed,the average comparison rate with reference genome was 96.72%;the genome coverage depth was 84.92%(at least one base covered)with an average coverage depth of 10X.Detection and annotation of SNP and InDel of the sample were completed,the genetic evolution and genetic diversity analysis were analyzed,and the native poplar populations were evaluated.The results of phylogenetic variation detection showed that phylogeny of Sect.Leuces was early,and it had a distant relationshipwith Sect.Aigeiros and Sect.Tacamahacain phylogeny,and there was interspecific gene flow in Sect.Tacamahaca.The present study will provide reference for the protection and utilization of native poplar.
