降香通脉汤对心肌缺血再灌注大鼠炎症因子及NLRP3/ASC/Caspase-1信号通路的影响 被引量:2

摘要 目的:观察降香通脉汤对心肌缺血再灌注大鼠炎症因子及NLRP3/ASC/Caspase-1信号通路的影响。方法:将50只SD大鼠随机分为假手术组、模型组及降香通脉高、中、低剂量组,每组10只,给药14天后,采用大鼠冠状动脉前降支结扎30 min再复流1 h的方法建立I/R模型。ELISA法检测大鼠血清指标,HE染色观察心肌形态学改变,Western blot法检测心肌组织NLRP3、ASC、Caspase-1蛋白表达。结果:与假手术组比较,模型组大鼠心肌梗死面积、血清LDH、AST、CK-MB、IL-1β及IL-6水平及NLRP3、Caspase-1、ASC表达均增高(P<0.05);与模型组比较,降香通脉组上述各指标明显降低(P<0.05),且高剂量组下降最明显。结论:降香通脉汤可以降低再灌注大鼠心肌梗死面积,能降低大鼠血清AST、CK-MB、LDH指标和炎症因子IL-1β和IL-6的水平。其机制可能是通过抑制NLRP3/ASC/Caspase-1信号通路对再灌注心肌起到保护作用。 Objective:To observe the effect of Jiangxiang Tongmai decoction on inflammatory factors and NLRP3/ASC/Caspase-1 signal pathway in rats with myocardial ischemia reperfusion.Methods:50 SD rats were randomly divided into sham operation group,model group and high-,medium-and low-dose groups of Jiangxiang Tongmai decoction,10 rats in each group.After 14 days of administration,I/R model was established by ligating the anterior descending branch of coronary artery of rats for 30 minutes and reflow for 1 hour.Serum indexes of rats were detected by ELISA.The changes of myocardial morphology were observed by HE staining.The protein expressions of NLRP3,ASC and Caspase-1 in myocardial tissue were detected by Western blot.Results:Compared with sham operation group,the area of myocardial infarction,the levels of serum LDH,AST,CK-MB,IL-1βand IL-6 and the expressions of NLRP3,Caspase-1 and ASC of rats in model group were increased(P < 0.05).Compared with model group,the above indexes in Jiangxiang Tongmai decoction group,especially the high-dose group,were decreased significantly(P<0.05).Conclusion:Jiangxiang Tongmai decoction can reduce the area of myocardial infarction in rats with reperfusion,the indexes of serum AST,CK-MB and LDH in rats and the levels of inflammatory factors IL-1βand IL-6 in rats with reperfusion.The mechanism may be that the inhibition of NLRP3/ASC/Caspase-1 signal pathway plays a protective effect on myocardium with reperfusion.
