摘要
采用酶底物法测定水中总大肠菌群。通过正交实验从样品放置室温时间、培养温度、培养时间和稀释用水等方面探讨酶底物法测定水中总大肠菌群的各种因素的影响。实验表明样品放置室温时间对水中总大肠菌群的测定的影响因素最大,第二为培养温度,第三为培养时间,影响最低的为稀释用水。酶底物法测定水中总大肠菌群的条件为:室温放置时间在20~23 min,培养温度在35.0~38.0℃,培养时间在24~28 h,采用稀释溶液(爱德士公司)、灭菌生理盐水(自配)来稀释样品。
The enzyme substrate method was used to determine the total coliform bacteria in water. The influence of various factors on the determination of total coliform bacteria in water by enzyme substrate method was discussed from the aspects of room temperature, culture temperature, culture time and dilution water. The experiment showed that the temperature of the sample was the most influential factor on the determination of total coliform bacteria in water. The second was culture temperature, the third was culture time, and the lowest was dilution water. The conditions for the determination of total coliform bacteria in water by enzyme substrate method were as follows: the incubation time was 20~23 min at room temperature, the incubation temperature was 35.0~38.0 ℃, and the incubation time was 24~28 h. The samples were diluted with diluted solution(IDEXX Co., Ltd) and sterilized saline(self-prepared).
