基于基因数据库分析GRB2在肾癌中的表达及其与患者预后的关系

EXPRESSION OF THE GROWTH FACTOR RECEPTOR-BOUND PROTEIN-2 GENE IN RENAL CARCINOMA AND ITS ASSOCIATION WITH PATIENT PROGNOSIS BASED ON GENE DATABASE ANALYSIS

目的探讨生长因子受体结合蛋白2(GRB2)基因在肾癌中的表达及其对患者预后的影响。方法挖掘ONCOMINE数据库中肾癌GRB2基因相关数据进行基因表达分析,结合在TCGA数据库中挖掘的GRB2基因在肾癌组织和正常组织中的表达差异,获得GRB2基因的表达与患者预后的关系。并通过STRING数据库,构建GRB2基因的蛋白质-蛋白质互作网络图。采取实时荧光定量PCR的方法检测6例肾癌组织及其癌旁正常组织中GRB2基因的相对表达量。结果与正常肾组织相比,GRB2基因在肾癌组织中的表达明显升高,差异具有统计学意义(t=8.21,P<0.05),GRB2基因在肾癌组织中的表达升高与肾癌患者预后相关(z=2.71,P<0.05)。进一步对GRB2相关的14个相互作用蛋白进行分析显示,GRB2主要参与了细胞的增殖和凋亡过程。肾癌组织中GRB2基因相对表达量显著高于其癌旁正常组织(t=6.98,P<0.001)。结论GRB2基因在肾癌组织中高表达,且其基因表达水平与肾癌患者的预后明显相关。

Objective To investigate the expression of the growth factor receptor-bound protein-2(GRB2)gene in renal carcinoma and its influence on prognosis.Methods The ONCOMINE database was used to obtain the data on the expression of the GRB2 gene in renal carcinoma,and the TCGA database was used to analyze the difference in the expression of the GRB2 gene between renal carcinoma tissue and normal tissue,as well as the association between the expression of the GRB2 gene and the prognosis of patients.The STRING database was used to construct the protein-protein interaction network of the GRB2 gene.Quantitative real-time PCR was used to measure the relative expression level of the GRB2 gene in six renal carcinoma tissue samples and corresponding adjacent normal tissue samples.Results The expression of the GRB2 gene in renal carcinoma tissue was significantly higher than that in normal renal tissue(t=8.21,P<0.05),and the increased expression of the GRB2 gene in renal carcinoma tissue was associated with the prognosis of renal carcinoma(z=2.71,P<0.05).Further analysis of the 14 GRB2-related interacting proteins showed that GRB2 was mainly involved in cell proliferation and apoptosis.The relative expression level of the GRB2 gene in renal carcinoma tissue was significantly higher than that in the corresponding adjacent normal tissue(t=6.98,P<0.001).Conclusion The GRB2 gene is highly expressed in renal carcinoma tissue,and its expression level is significantly associated with the prognosis of patients with renal carcinoma.
