食管鳞癌中miR-122-5P的表达及其临床病理学意义

Expression of miR-122-5P in Esophageal Squamous Cell Carcinoma and Its Clinicopathological Significance

目的探究食管鳞癌组织中miR-122-5P的表达、临床病理学意义及生物学行为。方法选取食管鳞癌患者74例,收集手术切除的食管鳞癌组织及其配对的癌旁正常食管组织。采用原位杂交技术检测miR-122-5P的表达,比较食管鳞癌组织与癌旁正常食管组织miR-122-5P表达差异,分析其表达及与肿瘤直径、病理分级、淋巴结转移等临床病理特征的关系。结果miR-122-5P在食管鳞癌中的阳性表达率35.1%(26/74)明显低于癌旁正常食管组织77.0%(57/74)(χ^(2)=26.363,P=0.000)。miR-122-5P与食管鳞癌的肿瘤部位、浸润深度表达差异有统计学意义(χ^(2)=16.662,P=0.000;χ^(2)=10.723,P=0.005)。食管鳞癌组织中miR-122-5P的表达水平与患者性别、年龄、病理类型、肿瘤直径、病理分级、脉管转移、神经转移及淋巴结转移差异均无统计学意义(P>0.05)。结论miR-122-5P在食管鳞癌组织和癌旁组织表达不同,食管鳞癌组织中表达下调,推测其可能具有抑癌基因的功能。

Objective To investigate the expression,clinicopathological significance and biological behavior of miR-122-5P in esophageal squamous cell carcinoma(ESCC).Methods 74 patients with ESCC were selected and their adjacent normal tissues were collected.In situ hybridization was used to detect the expression of miR-122-5P,and the difference of miR-122-5P expression between ESCC tissues and normal esophageal tissues adjacent to cancer was compared,and the relationship between miR-122-5P expression and clinicopathological features such as tumor diameter,pathological grade and lymph node metastasis was analyzed.Results The positive expression rate of miR-122-5P in ESCC(35.1%,26/74)was significantly lower than that in adjacent normal esophageal tissue(77.0%,57/74)(χ^(2)=26.363,P=0.000).miR-122-5P expression was significantly different in tumor site and invasion depth of ESCC(χ^(2)=16.662,P=0.000;χ^(2)=10.723,P=0.005).The expression level of miR-122-5P in ESCC showed no significant difference in gender,age,pathological type,tumor diameter,pathological grade,vascular metastasis,nerve metastasis and lymph node metastasis(P>0.05).Conclusion The expression of miR-122-5P is different in ESCC tissues and paracancer tissues,and the expression of miR-122-5P is down-regulated in ESCC tissues,suggesting that miR-122-5P may have the function of tumor suppressor gene.
