野鸢尾黄素对人乳腺癌细胞MCF7增殖、迁移和侵袭的影响

摘要 目的探讨野鸢尾黄素对人乳腺癌细胞MCF7增殖、侵袭和迁移的影响及其可能的作用机制。方法将MCF7细胞分为低剂量组、高剂量组和对照组。低、高剂量组分别加入40μmol/L和80μmol/L野鸢尾黄素,对照组不加任何药物。用CCK-8实验检测细胞增殖能力,用细胞划痕实验检测细胞的迁移能力,用Tranwell小室实验检测细胞的侵袭能力,用蛋白质印迹法检测β-catenin蛋白及p-GSK-3β-ser9的表达水平。结果干预48 h后,对照组和低、高剂量组的细胞增殖率分别为(100±4.00)%、(62.9±2.60)%、(32.2±6.25)%,迁移的距离分别为(392±7.32)μm、(272±17.45)μm和(92±5.36)μm,侵袭细胞数量分别为(130±3.33)个、(96±3.33)个和(66±2.66)个,β-catenin相对GAPDH的相对表达量分别为0.567±0.06、0.312±0.06和0.243±0.05,p-GSK-3β-ser9相对GAPDH的相对表达量分别为0.501±0.06、0.236±0.04和0.166±0.03。低、高剂量组的上述指标分别与对照组比较,以及高剂量组与低剂量组间比较,差异均有统计学意义(P<0.05)。结论野鸢尾黄素可能通过抑制Wnt/β-catenin信号通路,从而抑制MCF7细胞的增殖、迁移及侵袭。 Objective To observe the effect of tectorigenin on the proliferation,invasion,and migration of breast cancer cells MCF7 and its possible mechanism.Methods MCF7 cells were divided into three groups:control group,low-dose and high-dose groups,with no drugs,40μmol/L and 80μmol/L tectorigenin added,respectively.Cell proliferation was detected by CCK-8 assay,and cell migration was detected by cell scratch assay.Cell invasion was detected by Tranwell invasion assay,andβ-catenin protein expression was detected by western blot.Results After 48 h of intervention,the proliferation rates of cells in blank control group,and low-dose and high-dose groups were(100±4.00)%,(62.9±2.60)%,(32.2±6.25)%,and the migration distance was(392±7.32)μm,(272±17.45)μm,and(92±5.36)μm,respectively.The number of invasive cells was 130±3.33,96±3.33,and 66±2.66,and the relative expression of β-catenin was 0.567±0.06,0.312±0.06,and 0.243±0.05,respectively.Meanwhile,the relative expression of p-GSK-3β-ser9 was 0.501±0.06,0.236±0.04,and 0.166±0.03,respectively.There were statistically significant differences in the above indexes between the control group and the low-dose and high-dose groups(P<0.05),as well as between the low-dose group and high-dose group(P<0.05).Conclusion The expression of tectorigenin may inhibit the proliferation,migration and invasion of MCF7 cells through inhibiting Wnt/β-catenin signal pathway.
