摘要
铁死亡是新近提出来的一种铁依赖性、脂质活性氧累积独特的细胞程序性死亡方式,细胞发生铁死亡过程主要涉及有铁离子代谢紊乱、铁诱导通过不同途径影响谷胱甘肽过氧化物酶的活性状态,导致体内抗氧化防御系统失衡并触发大量脂质过氧化物的堆积。而铁积累是发生铁死亡的驱动因素,铁超载及脂质过氧化物累积共同介导骨骼肌再生和修复稳态的破坏,最终导致老年肌肉减少症的发生。但目前关于铁死亡如何参与骨骼肌系统的相关研究甚少,因此本文就铁死亡的发生机制及其与老年肌肉减少症的关系进行综述,旨在为进一步了解老年肌肉减少症发病机制和治疗该疾病的新靶点提供参考。
Ferroptosis is a newly proposed unique programmed cell death mode of iron dependence and lipid reactive oxygen species accumulation.The process of ferroptosis in cells mainly involves iron ion metabolism disorder and iron induction,which affects the activity of glutathione peroxidase through different ways,leading to the imbalance of antioxidant defense system in vivo and triggering the accumulation of a large number of lipid peroxides.Iron accumulation is a driving factor for ferroptosis.Iron overload and lipid peroxide accumulation jointly mediate the destruction of skeletal muscle regeneration and repair homeostasis,eventually leading to the occurrence of sarcopenia in the elderly.However,there are few studies on how ferroptosis participates in the skeletal muscle system.Therefore,this article reviews the mechanism of ferroptosis and its relationship with sarcopenia in the elderly,aiming to provide a reference for further understanding the pathogenesis of sarcopenia in the elderly and new targets for the treatment of the disease.
