金卷升板胶囊对模型大鼠血小板减少的改善作用

Effect of Jinjuan Shengban Capsules on the Improvement of Thrombocytopenia in Model Rats

目的 探讨金卷升板胶囊对模型大鼠血小板减少的改善作用。方法 将36只雄性SD大鼠随机分为空白对照组(A组,等体积生理盐水),模型组(B组,等体积生理盐水),咖啡酸组(C组,9 mg/kg),以及金卷升板胶囊高、中、低剂量组(D_(1)组、D_(2)组、D_(3)组,1.00,0.50,0.25 g/kg),各6只。腹腔注射环磷酰胺溶液(100 mg/kg),每日1次,连续3 d,以复制血小板减少大鼠模型,同时A组大鼠腹腔注射等体积生理盐水。在建模开始后第5天,各组大鼠予相应药物或生理盐水灌胃,每日1次,连续14 d。检测血小板计数(PLT)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(Fib);采用酶联免疫吸附法检测白细胞介素10(IL-10)、肿瘤坏死因子-α(TNF-α)的水平;称定脾脏质量并计算脾脏系数;采用苏木素-伊红染色,并在显微镜下观察脾脏组织病理形态。结果 与A组比较,B组大鼠PLT显著减少,PT,APTT,TT均显著延长,Fib和TNF-α水平均显著升高,IL-10水平显著降低,脾脏系数显著降低(P<0.01)。与B组比较,C组、D_(1)组、D_(2)组大鼠PLT显著增加,PT,APTT,TT均显著缩短,Fib及TNF-α水平均显著降低;C组、D_(1)组、D_(2)组、D_(3)组大鼠IL-10水平均显著升高,脾脏系数显著升高(P<0.01);D_(1)组、D_(2)组、D_(3)组大鼠脾脏组织的B淋巴细胞数量均增加,细胞核内的嗜碱性物质着色明显,细胞核完整度较好。结论 金卷升板胶囊可改善模型大鼠的血小板减少情况,其机制可能与升高IL-10水平及降低TNF-α水平和改善大鼠的免疫有关。

Objective To investigate the effect of Jinjuan Shengban Capsules on the improvement of thrombocytopenia in model rats.Methods Thirty-six male SD rats were randomly divided into the blank control group (group A,equal volume of normal saline),model group (group B,equal volume of normal saline),caffeic acid group (group C,9 mg/kg) and high-,medium-and lowdose groups of Jinjuan Shengban Capsules (group D_(1),group D_(2),group D_(3);1.00,0.50,0.25 g/kg),six rats in each group.The rats in groups B,C,D_(1),D_(2),and D_(3)were intraperitoneally injected with cyclophosphamide solution (100 mg/kg) once a day for 3 d to replicate the rat model with thrombocytopenia,while the rats in group A were given same volume of normal saline by intraperitoneal injection.On the 5th day after the start of modeling,rats in each group were given corresponding drugs or physiological saline by gavage once a day for 14 d.The platelet count (PLT),prothrombin time (PT),activated partial thromboplastin time (APTT),thrombin time (TT) and fibrinogen (Fib) were measured.The levels of interleukin-10(IL-10) and tumor necrosis factor-α(TNF-α) in rats were measured by the enzyme-linked immunosorbent assay (ELISA).The spleen mass was measured and the spleen coefficient was calculated.Hematoxylin-eosin (HE) staining was used for observing the histopathological morphology of spleen under the microscope.Results Compared with those in group A,the PLT was significantly lower,the PT,APTT and TT were significantly longer,the levels of Fib and TNF-α were significantly higher,the level of IL-10was significantly lower,while the spleen coefficient was significantly lower in group B (P<0.01).Compared with those in group B,the PLT was significantly higher,the PT,APTT and TT were significantly shorter,the levels of Fib and TNF-α were significantly lower in groups C,D_(1)and D_(2);the level of IL-10 was significantly higher,while the spleen coefficient was significantly higher in groups C,D_(1),D_(2)and D_(3)(P<0.01);the number of B lymphocytes in the spleen tissue of rats increased,the basophils in the nucleus were obviously stained,and the nuclear integrity was good in groups D_(1),D_(2)and D_(3).Conclusion Jinjuan Shengban Capsules can improve the thrombocytopenia of model rats,and its mechanism may be related to the increase of IL-10 level,the decrease of TNF-α level,and the improvement of immunity in rats.
