硬毛粗盖孔菌多糖分级醇沉及抗氧化活性研究 被引量:1

摘要 通过对硬毛粗盖孔菌进行液体发酵培养获得菌丝,以冻干菌丝为原料,采用超声辅助热水法提取多糖。用不同终浓度的乙醇溶液(40%、60%和80%)进行沉淀,分别得到(FCP-40、FCP-60和FCP-80)3个组分。同时,运用不同方法对其化学组成、结构特征和抗氧化活性进行了探究。结果表明,3个组分的多糖均含有核酸和蛋白质,其中FCP-40的总糖含量最高,FCP-80的蛋白质含量最高。不同醇沉组分多糖均含有较长侧链和较多分支且都是三螺旋多糖。3种多糖均具有剂量依赖性的抗氧化活性,且羟基(·OH)清除能力相较DPPH自由基清除能力而言效果更显著。 Mycelium was obtained by liquid fermentation culture of Funalia trogii,and polysaccharides were extracted by ultrasound-assisted hot water method using freeze-dried mycelium as raw material.Precipitation with ethanol solutions of different final concentrations(40%,60% and 80%)into three fractions(FCP-40,FCP-60 and FCP-80)respectively.The chemical composition,structural characteristics and antioxidant activity were also investigated using different methods.The results showed that all three polysaccharide fractions contained nucleic acids and proteins,with the highest total sugar content in the FCP-40 and the highest protein content in the FCP-80.All three polysaccharides are contain longer side chains and more branches and are triple helix polysaccharides.All three polysaccharides have dose-dependent antioxidant activity and are more effective in scavenging hydroxyl groups than DPPH radicals.
