壮医药线点灸对围绝经期大鼠血清IL-2、IFN-γ和IL4、IL10表达的影响

Effect of Zhuang Medicine Thread Moxibustion on the Expression of IL-2,IFN-γ,IL 4 and IL 10 in Serum of Perimenopausal Rats

目的观察壮医药线点灸对围绝经期大鼠血清白细胞介素(Interleukin,IL)-2、γ-干扰素(Interferon,FN-γ)、IL-4和IL-10表达变化的影响。方法通过化合物4-二氧化乙烯基环己烯(4-vinylcyclohexene dioxide,4-VCD)皮下注射制备围绝经期综合征(Perimenopausal syndrome,MPS)大鼠模型。采用阴道细胞学检查方法筛选出围绝经期模型大鼠并随机分为模型组、壮医药线点灸治疗组(药线组)和普通针刺治疗组(针刺组),每组10只,另设10只雌性大鼠作为正常组。治疗疗程为每日1次,连续6个月经周期,其中正常组和模型组常规饲养,无治疗。末次治疗24 h后,称取各组大鼠体质量,腹主动脉采血并处死大鼠,取出卵巢,HE染色法观察各组大鼠卵巢组织形态学变化,酶联免疫吸附测定法(Enzyme linked immunosorbent assay,ELISA)法测定各组大鼠血清IL-2、IFN-γ、IL-4和IL-10的含量。结果与模型组相比,药线组和针刺组能明显升高模型组大鼠血清中IL-2和IFN-γ的含量,降低模型组大鼠血清中IL-4和IL-10的含量(P<0.05),且药线组改善情况优于针刺组(P<0.05)。HE染色发现,药线组和针刺组卵巢组织中各级卵泡较模型组明显增多,间质未见明显纤维组织增生。结论壮医药线点灸疗法能显著提高围绝经期模型大鼠血清中IL-2和IFN-γ水平,降低IL-4和IL-10的水平,改善大鼠围绝经期的卵巢病理变化,其机制可能与调节机体内分泌-免疫系统有关。

Objective To observe the effect of Zhuang medicine thread moxibustion on the expression of serum IL-2,IFN-γ,IL-4 and IL-10 in perimenopausal rats.Methods The rat model of perimenopausal syndrome was established by subcutaneous injection of compound 4-VCD.The perimenopausal rats were randomly divided into model group,Zhuang medicine thread moxibustion treatment group(medicine thread group)and ordinary acupuncture group(acupuncture group),with 10 rats in each group,and 10 female rats as normal group.The treatment was once a day for 6consecutive menstrual cycles.24 hours after the last treatment,the body weight of rats in each group was weighed,blood was collected from abdominal aorta,and the ovaries were taken out.The histological changes of ovaries were observed by HE staining,and the contents of serum IL-2,IFN-γ,IL-4 and IL-10 were measured by ELISA.Results Compared with the model group,the drug thread group and acupuncture group could significantly increase the contents of IL-2 and IFN-γin the serum of the model group,and reduce the contents of IL-4 and IL-10 in the serum of the model group(P<0.05),and the improvement of the medicine thread group was better than that of the acupuncture group(P<0.05).He staining showed that the number of follicles in ovarian tissue of drug thread group and acupuncture group was significantly higher than that of model group,and no obvious fibrous tissue hyperplasia was found in stroma.Conclusion Zhuang medicine thread moxibustion can significantly increase the serum levels of IL-2 and IFN-γ,reduce the levels of IL-4 and IL-10 in perimenopausal rats,and improve the pathological changes of ovaries in perimenopausal rats,the mechanism may be related to the regulation of endocrine immune system.
