大麻二酚对羊膜细胞分泌炎症相关因子的影响

摘要 目的研究大麻二酚(CBD)对羊膜细胞分泌炎症相关因子的影响。方法体外培养WISH羊膜细胞,采用不同浓度CBD预处理后,再用脂多糖(LPS)刺激24 h。Western blot测定核因子κB(NF-κB)和p38的活性,定量PCR测定肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)mRNA的表达。酶联免疫吸附试验(ELISA)测定培养上清中前列腺素E_(2)(PGE_(2))和环氧化酶-2(COX-2)的水平。结果CBD处理组NF-κB的抑制因子IκB表达水平高于LPS组,p38磷酸化水平低于LPS组,细胞中IL-1β和TNF-α的mRNA表达也低于LPS组,差异均有统计学意义(P<0.05)。此外,CBD处理组中PGE_(2)和COX-2的水平也低于LPS组,差异有统计学意义(P<0.05)。结论CBD抑制LPS诱导WISH细胞分泌炎症相关因子。 Objective To study the effect of cannabinoid(CBD)on amniotic cells secretion of inflammatory related factors.Methods WISH cells were cultured in vitro,pretreated with CBD and stimulated with lipopolysaccharide(LPS)for 24 hours.The activities of nuclear factor-κB(NF-κB)and p38 were determined by Western blot.The mRNA of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were detected by quantitative PCR.The levels of prostaglandin-2(PGE_(2))and cyclooxygenase-2(COX-2)in the culture supernatant were determined by enzyme-linked immunosorbent assay(ELISA).Results The NF-κB inhibitor IκB expression in CBD group was higher than that in LPS group,p38 phosphorylation level was lower than that in LPS group,the mRNA levels of IL-1βand TNF-αwere also lower than those in LPS group,and the differences were statistically significant(P<0.05).In addition,the levels of PGE_(2) and COX-2 in CBD treatment group were also lower than those in LPS group,and the differences were statistically significant(P<0.05).Conclusion CBD inhibits the secretion of inflammatory related factors by amniotic cells induced by LPS.
