摘要
本文采用热水浸提法提取板栗种仁中的总糖,借助微滤—超滤—纳滤的膜分离组合工艺分离其中的寡糖成分,利用阴离子交换层析对粗寡糖进行纯化后得到中性板栗寡糖NOC。理化性质测试表明,寡糖NOC是重均分子量为670Da的四元寡糖,化学组成几乎不含蛋白质和糖醛酸,微观形貌呈现纤维状聚集形态。采用DPPH法测试了寡糖NOC的抗氧化能力,其对DPPH自由基的半数清除浓度EC50为5.01mg/mL,显示出较好的抗氧化活性。
In this paper, the total sugars in chestnut seed kernels were extracted by hot water extraction, and the oligosaccharide components were separated by a combined microfiltration-ultrafiltration-nanofiltration membrane separation process. The neutral chestnut oligosaccharide NOC was obtained after purification of the crude oligosaccharide by anion-exchange chromatography. Physical and chemical property tests showed that the oligosaccharide NOC is a tetrasaccharide with Mw of 670 Da, free of proteins and glyoxalates, has fibrous morphology. The antioxidant capacity of oligosaccharide NOC was tested by DPPH method, and the EC50 of half scavenging concentration for DPPH radicals was 5.01 mg/mL, showing a good antioxidant activity.
