摘要
目的:探讨盐酸去氢骆驼蓬碱(HH)抗恶性黑色素瘤细胞(B16F10)的体外作用机制。方法:CCK-8检测HH对B16F10细胞的生长抑制率,普通光学显微镜检测HH作用后B16F10的细胞形态变化,流式细胞术检测HH作用于B16F10的细胞周期的改变、细胞凋亡率以及活性氧的变化,荧光显微镜检测HH作用后B16F10细胞内自噬小体染色情况。结果:与阴性对照组相比,实验组B16F10的生长抑制率、细胞凋亡率显著上升(P<0.05),G2/M期比例升高(P<0.05),细胞内活性氧含量升高(P<0.05),细胞形态呈圆形或者不规则改变,细胞内自噬小体比例升高。结论:HH能够诱导恶性黑色素瘤细胞B16F10发生凋亡及自噬,并将细胞周期阻滞在G2/M期。
Objective:To investigate the mechanism of harmine hydrochloride(HH)against malignant melanoma cells(B16F10)in vitro.Methods:The growth inhibition rate of B16F10 cells by HH was detected by CCK-8,the morphological changes of B16F10 cells after HH were detected by ordinary light microscope,the changes of cell cycle,apoptosis rate and reactive oxygen species of B16F10 cells after HH were detected by flow cytometry,and the staining of autophagy bodies in B16F10 cells after HH was detected by fluorescence microscope.Results:Compared with the negative control group,the growth inhibition rate and apoptosis rate of B16F10 in the experimental group increased significantly(P<0.05),the proportion of G2/M phase increased(P<0.05),the content of intracellular reactive oxygen species increased(P<0.05),the cell morphology changed round or irregularly,and the proportion of intracellular autophagy increased.Conclusion:Harmine hydrochloride can induce apoptosis and autophagy of malignant melanoma cell line B16F10,and block the cell cycle in G2/M phase.
