组织蛋白酶D在脑卒中后炎性反应中的作用及研究进展

摘要 组织蛋白酶D是在溶酶体中起作用的天冬氨酸蛋白酶,并在细胞质中广泛地参与自噬这一过程,参与蛋白质的降解以及诱导细胞凋亡。脑卒中后的炎性反应在脑损伤中发挥重要作用,组织蛋白酶D参与自噬及脑卒中后炎症的级联反应。研究显示,组织蛋白酶D通过增强脑卒中后炎性反应而参与脑卒中后的病理损伤过程。组织蛋白酶D在脑卒中后损伤中的具体机制可以为进一步探索炎性脑损伤治疗措施奠定基础,组织蛋白酶D有望成为临床干预和治疗脑卒中的新靶点,但具体机制仍需进一步探究。本文从组织蛋白酶D的功能及结构、与自噬的关系、组织蛋白酶抑制剂、脑卒中病程中的研究进展方面进行综述。 Cathepsin D is an aspartate protease that functions in lysosomes and is extensively involved in autophagy in cytoplasm,protein degradation and apoptosis induction.Inflammatory response after stroke plays an important role in brain injury,and cathepsin D is involved in the cascade of autophagy and inflammation after stroke.It has been found that cathepsin D is involved in the process of pathological injury after stroke by enhancing the inflammatory response after stroke.The specific mechanism of cathepsin D in post-stroke injury may lay a foundation for further exploration of therapeutic measures for inflammatory brain injury.Cathepsin D is expected to become a new target for clinical intervention and treatment of stroke,but the specific mechanism still needs further exploration.This article reviews the function and structure of cathepsin D,the relationship between cathepsin D and autophagy,cathepsin inhibitor and the research progress in the course of stroke.
