四磨汤对肝脾气滞证功能性消化不良大鼠胃肠运动及血清GAS、MTL的影响 被引量:9

Effects of Si Mo Decoction on gastrointestinal motility and serum GAS and MTL in rats with functional dyspepsia with syndrome of liver-spleen Qi stagnation

摘要 目的探讨四磨汤治疗肝脾气滞证功能性消化不良的作用及可能机制。方法将36只SPF级Wistar大鼠随机分为空白组、模型组、莫沙必利组和四磨汤低、中、高剂量组,每组6只。除空白组外,其余组大鼠采用夹尾刺激法和不规则喂养法制备肝脾气滞证功能性消化不良模型。造模成功后,四磨汤低、中、高剂量组分别给予1.8 g/(kg·d)、2.4 g/(kg·d)、3.2 g/(kg·d)四磨汤灌胃,莫沙必利组给予莫沙必利0.305 mg/(kg·d)灌胃,空白组、模型组给等量生理盐水灌胃,均每日1次,连续灌胃14 d。灌胃结束后称量各组大鼠体重,采用酚红法测定胃排空率,采用碳粉法测定小肠推进速率,ELISA法检测血清胃动素(MTL)、胃泌素(GAS)水平。结果灌胃结束后,模型组大鼠体重、胃排空率、小肠推进率及血清中MTL、GAS水平均明显低于空白组(P均<0.05),四磨汤中、高剂量组和莫沙必利组大鼠体重、胃排空率、小肠推进率及血清中MTL、GAS水平均明显高于模型组(P均<0.05);四磨汤高剂量组大鼠体重与莫沙必利组比较差异无统计学意义(P>0.05),胃排空率及血清中MTL、GAS水平均明显高于四磨汤低、中剂量组和莫沙必利组(P均<0.05);四磨汤高剂量组及莫沙必利组大鼠小肠推进率明显高于四磨汤低剂量组(P均<0.05),四磨汤高剂量组与莫沙必利组比较差异无统计学意义(P>0.05)。结论四磨汤可增强肝脾气滞证功能性消化不良大鼠胃肠蠕动功能,机制可能与提高血清MTL、GAS水平有关。 Objective It is to investigate the effect and possible mechanism of action of Si Mo Decoction in the treatment of functional dyspepsia with syndrome of liver-spleen Qi stagnation.Methods Thirty-six SPF-grade Wistar rats were randomly divided into blank group,model group,mosapride group,Si Mo Decoction low,medium and high dose groups,6 rats in each group.Except for the blank group,all the rats in the other groups were prepared by tail pinch stimulation and irregular feeding to establish models of functional dyspepsia with syndrome of liver-spleen Qi stagnation.After successful modeling,1.8 g/(kg·d),2.4 g/(kg·d)and 3.2 g/(kg·d)of Si Mo Decoction were given to the low,medium and high dose groups of Si Mo Decoction,respectively,and 0.305 mg/(kg·d)of mosapride was given to the mosapride group,while equal amounts of saline were given to the blank group and model group,all once a day,continuously treated for 14 days.The gastric emptying rate and small intestine propulsion rate of the rats were observed after 14 days.After gavage,the rats in each group were weighed,and their gastric emptying rate was detected by phenol red method,their small intestine propulsion rate was detected by carbon method,and the levels of serum gastrin(MTL)and gastrin(GAS)were measured by ELISA.Results After gavage,the body mass,gastric emptying rate,small intestine propulsion rate and the levels of serum MTL and GAS of the rats in the model group were significantly lower than those in the blank group(all P<0.05),the body mass,gastric emptying rate,small intestine propulsion rate and the levels of serum MTL and GAS of the rats in the Si Mo Decoction medium and high dose groups and mosapride group were significantly higher than those in the model group(all P<0.05).There was no statistically significant difference in body weight between the high dose of Si Mo Decotion group and mosapride group(P>0.05),but the gastric emptying rate and the levels of serum MTL and GAS of the rats in the Si Mo Decoction high dose group were significantly higher than those in the Si Mo Decoction medium and low dose groups and mosapride group(all P<0.05).The small intestine propulsion rate of the rats in the Si Mo Decoction high dose group and mosapride group were significantly higher than those in the Si Mo Decoction low dose group(all P<0.05),there was no significant difference between the Si Mo Decoction high dose group and mosapride group(P>0.05).Conclusion Si Mo Decoction can enhance the gastrointestinal motility of rats with functional dyspepsia with syndrome of liver-spleen stagnation,and the mechanism may be related to up-regulating the levels of serum MTL and GAS.
