精准医学时代治疗外周T细胞淋巴瘤的新型药物研究进展

Advances in novel drugs for peripheral T-cell lymphoma in the precision medicine era

外周T细胞淋巴瘤(peripheral T-cell lymphoma,PTCL)具有恶性程度高、分子异质性强、临床预后极差等特点。由于缺乏对PTCL病因及发病机制的认识,一线治疗进展长期停滞,PTCL患者的五年总生存率不足30%,临床诊疗面临巨大挑战。精准医学概念的提出及高通量多组学测序的发展推动了分子靶点的发现和药物的研发,多种新型药物在PTCL的临床治疗中展现出较好的抗肿瘤效果及耐受性,包括二氢叶酸还原酶抑制剂、组蛋白去乙酰化酶抑制剂、单克隆抗体及抗体偶联药物、信号通路抑制剂等。本文就精准医学时代下PTCL临床治疗中新型药物的研究进展进行综述。

Peripheral T-cell lymphoma(PTCL)has been characterized by high malignancy,high molecular heterogeneity and extremely poor prognosis.Due to lack of understanding of etiology and pathogenesis,the progress of firstline therapy in PTCL has remained stagnant for several years.The 5-year overall survival for PTCL patients is less than 30%and the clinical treatment of PTCL remains a massive challenge for human health.With the development of precision medicine and high-throughput multi-omics sequencing,numerous novel molecule-targeted drugs have been emerging with great therapeutic potential and tolerability,including dihydrofolate reductase inhibitors,histone deacetylase inhibitors,monoclonal antibodies,antibody drug conjugates,signaling pathway inhibitors and so on.In this review,we shall summarize the recent advances in novel drugs for the treatment of peripheral T-cell lymphoma in the precision medicine era.
