摘要 目的探讨结肠肿瘤组织中长链非编码RNA(long NON-Coding RNA,lncRNA)NONHSAT017458的表达情况及潜在临床意义。方法运用高通量测序技术,建立基因与lncRNAs差异表达谱。通过基因本体(GO)注释分析及京都基因和基因组百科全书(KEGG)通路富集分析,对差异表达的lncRNA NONHASAT017458进行功能注释。采用定量反转录聚合酶链反应法(qRT-PCR)对10例结肠癌及腺瘤患者结肠肿瘤及瘤旁正常组织检测lncRNA NONHSAT017458的表达,分析其表达量与患者临床病理特征的关系。结果筛选出表达差异明显的lncRNA NONHSAT017458,通过qRT-PCR方法在结肠癌中验证结果一致。与正常癌旁组织相比,结肠癌组织中lncRNA NONHSAT017458呈现表达升高,差异有统计学意义(P<0.05)。但与正常肿瘤旁组织相比,结肠腺瘤组织中lncRNA NONHSAT017458,差异无统计学意义(P>0.05)。lncRNA NONHSAT017458的表达与结肠癌患者年龄、性别、肿瘤大小、TNM分期无明显相关性(均P>0.05)。结论lncRNA NONHSAT017458在结肠癌肿瘤组织中表达上调,其可能作为临床中新型的潜在的诊断和治疗靶点。 Objective To investigate the expression of Long non-coding RNA(lncRNA)NONHSAT017458 in colon tumors and its potential clinical significance.Methods High-throughput sequencing was used to establish the differential expression profiles of genes and lncRNAs.The differentially expressed lncRNA NONHASAT017458 was functionally annotated by Gene Ontology(GO)annotation analysis and Kyoto Encyclopedia of genes and genomes(KEGG)pathway enrichment analysis.The expression of lncRNA NONHSAT017458 was detected by quantitative reverse transcription polymerase chain reaction(qRT-PCR)in 10 colon cancer and adenoma patients.The relationship between the expression level and the clinicopathological features of the patients was analyzed.Results lncRNA NONHSAT017458 was screened and the results were consistent by qRT-PCR in colon cancer.LncRNA NONHSAT017458 expression was elevated in colon cancer tissues compared with normal control tissues(P<0.05).There were no statistical significantly differences in lncRNA NONHSAT017458 expression in colon adenoma tissues compared with normal control(P>0.05).The expression of lncRNA NONHSAT017458 was not correlated with age,sex,tumor size and TNM stage of colon cancer patients(all P>0.05).Conclusion The expression of lncRNA NONHSAT017458 is upregulated in colon cancer tissues,and it may become a new potential diagnostic and therapeutic target in clinic.
