摘要
目的:研究广西HIV-1独特重组型毒株(URFs)的基因重组特点,为广西HIV-1疫情防控提供科学依据。方法:对1例pol区基因序列初步判断为URF的HIV-1感染者的血液样本进行RNA提取、巢式PCR扩增全长基因并测序,从PubMed等数据库检索2012—2022年发表的有关广西URFs近似全长基因组的文章及对应的序列,构建系统进化树,并进行重组模式、断点分析和耐药突变位点分析。结果:测序获得1例长度约8.9 kb的URF,该URF是以CRF08_BC为骨架插入CRF01_AE片段构成的二代重组毒株。将该URF与数据库中检索到的12例广西URFs共同分析,发现广西URFs主要来源于性传播途径感染者和静脉吸毒感染者。所有URFs均包含CRF01_AE片段,以01_AE/07_BC重组比例最高(69.2%,9/13),1例URF含有主要耐药位点。结论:广西已鉴定的URFs数量较少,以01_AE/07_BC重组为主,应加强对URFs的监测和鉴定,以全面了解广西HIV-1重组毒株的特点。
Object:To investigate the recombinant characteristics of HIV-1 unique recombinant forms(URFs)identified in Guangxi from 2012 to 2022,so as to provide a scientific basis for HIV-1 interventions in Guangxi.Methods:RNA isolation,nest-PCR and sequencing of near full-length genes were performed on the blood samples of an HIV-1 positive individual whose pol region gene sequence was identified as URF.The articles and near full-length sequence of URFs identified in Guangxi between 2012 and 2022 were retrieved from PubMed and other databases to construct phylogenetic tree and perform recombination pattern,breakpoint analysis and drug resistance mutation site analysis.Results:One URF with a length of about 8.9 kb was obtained by sequencing and it was a second-generation recombinant form composed of CRF08_BC as skeleton inserted into CRF01_AE fragment.By jointly analyzing this URF and 12 Guangxi URFs retrieved from the database,it was found that the URFs in Guangxi were mainly from sexually transmitted infections and intravenous drug users.All URFs comprised CRF01_AE fragment,and the recombinant proportion of 01_AE/07_BC was the highest(69.2%,9/13).One URF contained major drug resistance sites.Conclusion:The number of identified URFs in Guangxi is relatively small,most of which are 01_AE/07_BC recombination.The monitoring and identification of URFs should be strengthened to fully understand the characteristics of HIV-1 recombinant forms in Guangxi.
