益肾达络饮治疗多发性硬化的网络药理学机制研究 被引量:1

摘要 目的通过网络药理学广泛地挖掘益肾达络饮治疗多发性硬化的潜在机制。方法本研究通过检索中医药整合药理学研究平台TCMIP v2.0获取部分中药活性成分与靶点,未收录的中药利用基于结构相似度的方案预测获取活性成分以及靶点;通过TCMIP v2.0和GeneCards数据库获得疾病基因靶点;得到共有基因集后进行富集,分析潜在的治疗机制。结果本研究共得到益肾达络饮的活性成分488种,潜在治疗多发性硬化靶点795个,MCODE算法筛选得到核心靶点72个。DO富集结果显示共有基因集主要富集于神经系统疾病,如富集最高的脑病,以及富集显著的阿尔茨海默病、癫痫、偏头痛和痴呆。GO富集结果显示共有基因集主要富集在神经递质代谢相关和氨基酸合成相关生物过程。KEGG富集结果多与神经系统疾病密切相关,如阿尔茨海默病,神经活性受体与配体相互作用通路,以及朊病毒。核心靶点富集分析结果显示多富集在氨基酸代谢、氧化磷酸化等,与共有基因集富集结果在一定程度上具有一致性。结论益肾达络饮可能通过调节氨基酸代谢、氧化磷酸化等来达到保持中枢神经系统的稳定,从而治疗多发性硬化。 Objective:To explore the potential mechanism of Yishen Daluo Decoction in the treatment of multiple sclerosis through network pharmacology.Methods:Active ingredients and targets of TCM were obtained by searching the research platform of integrated pharmacology of TCM,TCMIP v2.0.The active ingredients and targets of unincluded TCM were predicted by using the scheme based on structural similarity.Disease gene targets were obtained by TCMIP v2.0 and GeneCards database.After obtaining the common gene sets,enrichment analysis was performed to analyze the potential therapeutic mechanisms.Results:A total of 488 kinds of active ingredients and 795 potential therapeutic multiple sclerosis targets in Yishen Daluo Decoction were obtained;72 core targets were screened by MCODE algorithm.DO enrichment results showed that the common gene set was mainly enriched in neurological diseases,such as the highest concentration of encephalopathy,as well as the significant concentration of Alzheimer′s,epilepsy,migraine and dementia.GO enrichment results showed that the common gene set was mainly enriched in neurotransmitter metabolism-related and amino acid synthesis-related biological processes;KEGG enrichment results were closely related to neurological diseases,such as Alzheimer’s disease,neuroactive receptor-ligand interaction pathways,and prions.The results of core target enrichment analysis showed that most enrichments were in amino acid metabolism,oxidative phosphorylation,etc.,and the results were consistent with the results of common gene set enrichment to a certain extent.Conclusion:Yishen Daluo Decoction may maintain the stability of the central nervous system by regulating amino acid metabolism and oxidative phosphorylation.
