摘要
建立管花肉苁蓉提取物中四种苯乙醇苷(松果菊苷、管花苷A、毛蕊花糖苷、异毛蕊花糖苷)的近红外光谱定量分析模型,以快速评价管花肉苁蓉提取物的质量。以HPLC法测定管花肉苁蓉提取物中四种苯乙醇苷的含量作为参考值,采用近红外光谱法(NIRS)结合偏最小二乘法(PLS)建立管花肉苁蓉提取物中四种苯乙醇苷成分的定量分析模型,并对模型进行验证。结果表明:松果菊苷、管花苷A、毛蕊花糖苷、异毛蕊花糖苷定量分析模型的最佳光谱预处理方法分别为:SNV+SG+FD、MSC+FD、SNV+SD、MSC+SD,校正相关系数(R2c)分别为0.9937、0.9945、0.9954、0.9948;预测相关系数(R2p)分别为0.9094、0.9222、0.8812、0.8529;校正均方根偏差(RMSEC)分别为0.3681、0.0376、0.1944、0.0524;预测均方根偏差(RMSEP)分别为1.426、0.1448、1.002、0.2821。近红外模型预测值与真实值(HPLC测定)无显著性差异。该方法操作简便、准确、分析速度快、无污染,可用于管花肉苁蓉提取物中四种苯乙醇苷含量的快速测定。
To establish the near-infrared spectroscopy quantitative analysis models of four Phenylethanol glycosides(echinacoside,tubuloside A,acteoside,Isoacteoside)in the extract of Cistanche tubulosa,so as to quickly evaluate the quality of the extract of Cistanche tubulosa.The chemical reference valuesof four Phenylethanol glycosides in the extract of Cistanche tubulosa were simultaneously determined by HPLC.The near-infrared spectroscopy(NIRS)combined with partial least squares(PLS)method was used to estabilish a quantitative analysis model for predicting the content of four Phenylethanol glycosides in Cistanche tubulosa extract.The best spectral parameters of model of quantitative analysis models of echinacoside,tubuloside A,acteoside,Isoacteoside were SNV+SG+FD,MSC+FD,SNV+SD and MSC+SD.The correlation coefficients set(R2c)were 0.9937,0.9945,0.9954 and 0.9948.The correlation coefficients of predictation set(R2p)were 0.9094,0.9222,0.8812 and 0.8529.The root mean square error of calibration(RMSEC)were 0.3681,0.0376,0.1944 and 0.0524.The root mean square error of predication(RMSEP)were1.426,0.1448,1.002 and 0.2821.There was no significant difference between the predicted value of near infrared model and the real value(determined by HPLC).This method is simple,accurate,fast and pollution-free.It can be used for the rapid determination of four Phenylethanol glycosides in the extract of Cistanche tubulosa.
