槲皮素和木犀草苷促进皮肤伤口愈合机制的实验研究

Experimental study on the mechanism of quercetin and luteolin-7-glucoside in promoting skin wound healing

摘要 目的探究槲皮素、木犀草苷对皮肤伤口再上皮化的影响及机制。方法以不同浓度的槲皮素、木犀草苷单独或联合处理小鼠全层皮肤缺损伤口,隔天观察伤口愈合情况。分别用苏木素_伊红染色和免疫组织化学染色观察伤口边缘皮肤再生情况、细胞增殖及表皮干细胞标志分子、β-catenin、c-Mye、Sox2表达;采用RT-PCR检测Sox2在表皮干细胞的表达。结果槲皮素、木犀草苷对小鼠能促进皮肤伤口愈合,增加伤口周围新生皮肤表皮层厚度,增加表皮基底层中增殖细胞和表皮干细胞数量以及β-catenin.c-Mye和Sox2表达;在体外能促进表皮干细胞表达Sox2。槲皮素和木犀草昔合用对伤口愈合的促进作用具有相加效应。结论槲皮素、木犀草苷通过上调β-catenin、c-Mye、Sox2表达促进表皮干细胞增殖,从而促进伤口愈合,两者合用具有相加效应。 Objective To investigate the effect and mechanism of quercetin and luteolin-7-glucoside on re-epithelialization of skin wound.Methods The full-thickness skin defect wounds of mice were treated with quercetin and luteolin-7-glucoside of different concentrations alone or jointly,and the wound healing was observed every other day.Hematoxylin eosin staining and immunohistochemical staining were used to observe the skin regeneration,cell proliferation and epidermal stem cell marker moleculesβ-Catenin,c-Mye,Sox2 expression.The expression of Sox2 in epidermal stem cells was detected by RT-PCR.Results The quercetin and luteolin-7-glucoside can promote skin wound healing,increase the thickness of new skin epidermis around the wound,increase the number of proliferating cells and epidermal stem cells in the epidermal basal layerβ-Catenin,c-Myc and Sox2 expression.It can promote the expression of Sox2 in epidermal stem cells in vitro.The combination of quercetin and luteolin has an additive effect on wound healing.Conclusion The quercetin and luteolin-7-glucoside promote the proliferation of epidermal stem cells by upregulating the expression ofβ-catenin,c-Mye and Sox2,which in turn accelerates wound healing.The combination of quercetin and luteolin-7-glucoside has an additive effect on wound healing.
