不同贮藏条件对木荷花粉活力的影响

摘要 研究以木荷Schima superba无性系清远101花枝及花朵为试验材料,采用TTC染色鉴定法,研究不同培养液培养木荷离体花枝的花粉活力和不同贮藏温度、湿度和堆积高度的花粉活力变化规律。研究表明:5 d短期内,MS或10%蔗糖有利于木荷花枝培养,花枝中花粉活力较高;4℃以下低温,70%空气湿度和1.5 cm低堆积高度更有利于花药中的花粉保存。结论:花枝在MS和10%蔗糖培养液中持续培养,短时间内具有较高的花粉活力,能满足木荷授粉需要;常温下花粉保存在70%中等湿度或较低堆积高度能维持较高的活性;木荷花粉的长期保存需要低温或超低温进行保存。 The pollen viability of isolated flowering branches and flowers of Schima superba asexual line Qingyuan 101 was used as test materials,and the TTC staining identification method was used to study the pollen viability of isolated flowering branches of S.superba cultured in different culture solutions and the change patterns of pollen viability at different storage temperatures,humidity and stacking heights.The results showed that:In the short term of 5 days,MS or 10%sucrose was beneficial to the flower branch culture of S.superba,and the pollen activity in the flower branch was higher;Low temperature below 4℃,70%humidity and 1.5 cm low stacking height are more conducive to the preservation of pollen in anthers.Conclusion:flowering branches of MS and 10%sucrose culture solution continuously cultivated with high pollen viability for a short period of time can meet the pollination needs of S.superba;pollen preservation at 70%medium humidity or low stacking height at room temperature can maintain high activity;long-term preservation of S.superba pollen requires low or ultra-low temperature for preservation.
