联合胶体金RNA非扩增等温检测技术的研究

摘要 目的在本研究中,我们创造性地提出将胶体金技术联合RNA非扩增等温检测技术,进行病原体核酸检测。方法通过对目的基因的DNA和RNA分别分析,进行探针设计并进行DEMBA检测,建立胶体金技术联合RNA非扩增等温检测技术,并对特异性和灵敏度进行评估。结果该方法的检测时间在15分钟以内,并且具有高灵敏度和特异性,该方法对于单链核酸的检测具有可行性和可靠性。结论该反应体系简单,操作简单,检测时间极短,具有应用推广价值。 Objective In this study,we creatively proposed to combine colloidal gold technology with RNA non amplification isothermal detection technology for pathogen nucleic acid detection.Methods By analyzing the DNA and RNA of the target gene,we designed the probe and performed DEMBA detection.The combine colloidal gold technology with RNA non amplification isothermal detection technology was established,and its specificity and sensitivity was evaluated.Results The detection time of this method was less than 15 min with high specificity and sensitivity.It is proved that this method is feasible and reliable for the detection of single stranded nucleic acids.Conclusion The reaction system is simple,the operation is simple,the detection time is very short,and it has the value of application and popularization.
