摘要 目的通过生物信息学方法研究细胞周期素依赖性激酶1(Cyclin-dependent kinase 1,CDK1)在肝癌(Hepatocellular carcinoma,HCC)组织中的表达差异,探索其在肝癌预后和免疫细胞浸润间的作用。方法从GEO数据库下载肝癌数据集,使用R软件分析差异基因,并取交集基因;通过GO和KEGG分析富集通路;使用STRING和cytoscape构建差异基因互作网络,结果可视化获取核心基因CDK1;UALCAN和GEPIA数据库分析肝癌中CDK1的表达水平,及其与临床特征的关系;HPA数据库分析CDK1蛋白在肝癌组织和正常组织的表达差异;通过Kaplan-Meier Plotter分析CDK1表达水平与HCC预后的关系;TIMER数据库分析CDK1与HCC肿瘤免疫细胞浸润、免疫细胞表面标记物的相关性。结果3个肝癌数据集筛选出共有的347个差异基因;KEGG通路显示差异基因主要参与细胞周期通路;与正常组织相比,CDK1基因在HCC组织中表达水平升高;CDK1高表达与肿瘤分期、年龄均有相关性;生存分析结果表明CDK1基因高表达患者生存时间明显低于低表达患者。CDK1基因与HCC中多种免疫细胞浸润水平及其表面标记物显著相关。结论CDK1基因在HCC中表达上调,高表达的CDK1肝癌患者预后不良,并且影响HCC免疫浸润水平。CDK1基因可能为HCC新的免疫治疗靶点。 Objective To study the differential expression of Cyclin-dependent kinase 1(CDK1)gene in Hepatocellular carcinoma(HCC)tissues through bioinformatics method,and further explore its role in the prognosis of liver cancer and immune cell infiltration.Methods The liver cancer data set was downloaded from GEO database,and the differential genes were analyzed using R software,and the intersection genes were taken.The enrichment pathway was analyzed by GO and KEGG.The differential gene interaction network was constructed using STRING and cytoscape,and the core gene CDK1 was obtained.UALCAN and GEPIA databases were used to analyze the expression level of CDK1 in liver cancer and its relationship with clinical features.The expression difference of CDK1 protein between liver cancer tissue and normal tissue was analyzed by HPA database.Kaplan-Meier Plotter was used to analyze the relationship between CDK1 expression level and HCC prognosis.The correlation between CDK1 and immune cell infiltration and immune cell surface markers of HCC tumor was analyzed by TIMER database.Results A total of 347 differential genes were screened from 3 liver cancer data sets.KEGG pathway showed that differential genes were mainly involved in cell cycle pathway.Compared with normal tissues,the expression level of CDK1 gene was increased in HCC tissues.The high expression of CDK1 was correlated with tumor stage and age.Survival analysis showed that the survival time of patients with high CDK1 gene expression was significantly lower than that of patients with low CDK1 gene expression.CDK1 gene was significantly correlated with the levels of multiple immune cell infiltration and its surface markers in HCC.Conclusions Our study demonstrated that CDK1 gene expression is up-regulated in HCC,overexpression of CDK1 result in a poor prognosis,and affects the level of HCC immune infiltration.CDK1 gene may be a new immunotherapy target for HCC.
