摘要 驱动蛋白超家族蛋白(KIFs)最早是从鱿鱼轴突中分离,并被确定为真核生物中普遍存在的轴突运输动力分子。它们都具有微管依赖性和ATP活性运动电位,可利用ATP水解产生的能量沿着微管运动。目前已经明确,KIFs在细胞内细胞器、大分子、囊泡以及有丝分裂的染色体运输中发挥重要作用。在有丝分裂过程中,KIFs通过精准调控作用时间而发挥作用,然而在肿瘤细胞中KIFs失调可能导致细胞生长不受控制,从而参与了肿瘤发生及进展。此外,许多KIFs与化疗药物的耐药性以及多种肿瘤的不良预后相关。本文旨在总结分析KIFs在肿瘤领域的研究进展,并讨论其在肿瘤治疗方面的潜在价值。 Kinesin superfamily proteins(KIFs)were first isolated from squid axons,and identified as kinetic molecules for axonal transport prevalent in eukaryotes.They all share a highly conserved motor domain,and they also have microtubule-dependent and ATP-active motor potentials,thereby using energy generated by ATP hydrolysis to move along microtubules.It is now well established that KIFs play important roles in intracellular organelles,macromolecules,vesicles,and mitotic chromosome transport.During mitosis,KIFs function by precisely regulating the timing of action,however,dysregulation of KIFs in tumor cells may lead to uncontrolled cell growth,thus participating in tumorigenesis and progression.In addition,many KIFs are associated with resistance to chemotherapeutic agents and poor prognosis in a variety of tumors.This article aims to summarize and analyze the research progress of KIFs in the field of oncology and discuss their potential value in tumor therapy.
