摘要
【目的】探明两类百合不同发育时期的腺苷二磷酸葡萄糖焦磷酸化酶(AGPase)活性、AGPase基因表达量与淀粉含量的相关性。【方法】使用qPCR、连续监测法、蒽酮法分别测定材料AGPase基因的表达量、酶活性及淀粉含量,再对比两类百合的异同,以得到两类百合AGPase基因的表达量、酶活性与淀粉含量各自的特点及各因素间的相关性。【结果】所有克隆到的AGPase基因都具有PLN02241家族蛋白结构特征;定量结果表明,两类百合AGPase大亚基基因表达模式相同,总体趋势类似。小亚基基因表达模式存在差异,且小亚基基因的表达量低于大亚基基因;淀粉及酶活性测定结果表明,两类百合总体趋势一致。相关性分析表明,AGPase小亚基基因的表达量、AGPase酶活性与淀粉含量在鳞片中高度线性相关,且叶片中的酶活性与鳞片中的淀粉含量也高度线性正相关。【结论】两类百合不同时期的淀粉含量及酶活性变化趋势相同,但同一发育时期不同类型百合的淀粉含量及酶活性存在差异。两类百合大小亚基基因的表达量在鳞茎不同时期的表达模式相似,但同一发育时期不同类型百合的基因表达量存在差异。AGPase小亚基基因的表达量与淀粉含量正相关。
【Objective】The present paeper aimed to explore the correlation of the enzyme activity of ADPG pyrophosphorylase(AGPase),the gene expression of AGPase with the starch content on two kinds of lilies at different developmental stages.【Method】qPCR,continuous reaction method and anthrone method were used to determine the gene expression,enzyme activity and starch content.The similarities and differences between the two kinds of lilies were compared to obtain the characteristics of AGPase expression,enzyme activity and starch content and the correlation between various factors.【Result】All the cloned AGPase genes were the structural characteristics of PLN02241 family proteins.The quantitative results showed that the expression patterns of AGPase large subunits gene were the same and the overall trend was similar.The expression patterns of small subunits gene were different.In addition,the expression of small subunit gene was lower than that of large subunit gene;The determination results of starch content and enzyme activity showed that the overall trend of the two kinds of lilies was the same.Correlation analysis showed that the expression of AGPase small subunit gene and AGPase enzyme activity were correlated with starch content in scales highly linearly,and the enzyme activity in leaves was also highly linearly positively correlated with starch content.【Conclusion】The starch content and enzyme activities of the two types of lilies had the same trend in different periods,but there were differences in the starch content and enzyme activities of different types of lilies at the same period.The expression patterns of large and small subunits gene of the two types of lilies were similar at different stages of bulb,but there were differences in the expression levels of different types of lilies at the same developmental stage.The expression of AGPase small subunit gene was positively correlated with starch content.
