内蒙古绒山羊血清生化指标、电解质浓度和热休克蛋白基因表达的季节性变化研究

Seasonal variation of blood biochemical, electrolyte concentrations and heat shock protein gene expression in Inner Mongolia cashmere goats

摘要 为了揭示内蒙古绒山羊血清生化指标、电解质浓度和热休克蛋白(HSPs)基因表达的季节性变化规律,试验选择11只全年放牧的2周岁内蒙古绒山羊母羊,在一年内每月颈静脉采集血液样品,测定血清生化指标、电解质浓度、激素水平和外周血单个核细胞HSPs基因的相对表达量。同时,记录放牧地环境温度、相对湿度、光照周期、降水量和天气情况,分析气象参数的全年变化规律。结果表明:血清促肾上腺皮质激素和皮质醇浓度及外周血单个核细胞中HSP60和HSP90基因表达量呈现明显的季节性变化规律,表现为冬夏季节较高,春秋季节较低。血清生化指标和电解质水平则未呈现一致性规律,具体表现为血清胆红素浓度秋季较低,血清镁浓度则秋季较高,血清肌酐和钾浓度夏季较低,血清氯浓度则夏季较高,血糖和血清磷浓度则为5月份最高,血清尿素浓度为冬夏季节较高,春秋季节较低。但血清钠浓度及外周血单个核细胞中HSP70和HSP110基因表达量未呈现季节性变化规律。说明季节性环境变量对内蒙古绒山羊血清生化指标和电解质浓度有影响,并促进了与适应过程有关的激素分泌和HSP基因表达。 The present study was conducted to reveal seasonal variation of blood biochemical, electrolyte concentration and heat shock protein(HSPs) gene expression in Inner Mongolia cashmere goats. A total of eleven 2-year-old grazing Inner Mongolia cashmere goats were selected, and blood samples were collected from the jugular vein every month for one year. Serum biochemical indexes, electrolyte concentrations, hormone levels and the relative expression of HSP genes in peripheral blood mononuclear cells were determined. Meanwhile, the ambient temperature, relative humidity, photoperiod, precipitation data and weather conditions were collected in grazing land, and the monthly variation of meteorological parameters were analyzed. The results showed that the concentrations of serum adrenocorticotropic hormone and cortisol and the expressions of HSP60 and HSP90 genes in peripheral blood mononuclear cells showed obvious seasonal changes, which were higher in winter and summer, but lower in spring and autumn. Serum biochemical and electrolyte levels did not show a consistent tendency. Specifically, the serum bilirubin concentration was lower in autumn, but the serum magnesium concentration was higher in autumn. The serum creatinine and potassium concentrations were lower but serum chloride concentration was higher in summer. Blood glucose and serum phosphorus concentrations were the highest in May. Serum urea concentration was higher in winter and summer, but lower in spring and autumn. However, the serum sodium concentration and the expressions of HSP70 and HSP110 genes in peripheral blood mononuclear cells did not show seasonal variation. The results indicated that seasonal environmental variables affected blood biochemical and electrolyte concentrations in Inner Mongolia cashmere goats, and promoted hormone secretion and HSP gene expression related to the adaptation process.
