摘要
为探究miR-106a对牦牛卵泡颗粒细胞(GCs)增殖和激素分泌的影响,本试验通过体外分离培养牦牛卵泡GCs,分别转染miR-106a模拟物(miR-106a mimics)和miR-106a抑制剂(miR-106a inhibitors),利用CCK-8法检测GCs细胞增殖,ELISA法分别检测雌二醇(E_2)和孕酮(P_4)激素分泌情况。结果显示,与对照组相比,24、48、72和96 h时,miR-106a mimics组和miR-106a inhibitors组牦牛GCs的细胞增殖受到极显著抑制(P<0.01)。与对照组相比,GCs转染48 h时,miR-106a mimics组E_2分泌量极显著升高,而miR-106a inhibitors组E_2分泌量极显著降低(P<0.01);GCs转染72 h时,miR-106a mimics组和miR-106a inhibitors组E_2分泌量均极显著降低(P<0.01);GCs转染48 h时,miR-106a mimics组和miR-106a inhibitors组P_4分泌量均极显著降低(P<0.01);而GCs转染72 h时则结果相反,miR-106a mimics组和miR-106a inhibitors组P_4分泌量均极显著升高(P<0.01)。结果表明,miR-106a转染牦牛GCs后对细胞增殖和激素分泌具有影响,为其在卵巢卵泡中的研究提供了数据支持。
In order to assess the effect of miR-106a on the proliferation and hormone secretion of yak follicular granulosa cells(GCs),this study isolated and cultured yak follicular GCs in vitro,transfected miR-106a mimics and miR-106a inhibitors,respectively,examined the proliferation of GCs by CCK-8,and detected the secretion of estradiol(E 2)and progesterone(P 4)by ELISA.The results showed that compared with the control group,at 24 h,48 h,72 h and 96 h,the cell proliferation of yak GCs in miR-106a mimics group and miR-106a inhibitors group was significantly inhibited(P<0.01).Compared with the control group,at 48 h after GCs transfection,the secretion of E 2 in miR-106a mimics group was significantly increased,while that in miR-106a inhibitors group was significantly decreased(P<0.01);at 72 h after GCs transfection,the E 2 secretion in miR-106a mimics group and miR-106a inhibitors group decreased significantly(P<0.01);at 48 h after GCs transfection,P 4 secretion in miR-106a mimics group and miR-106a inhibitors group decreased significantly(P<0.01),in contrast,the P 4 secretion in miR-106a mimics group and miR-106a inhibitors group was significantly increased at 72 h after GCs transfection(P<0.01).Thus,miR-106a demonstrates some effect on cell proliferation and hormone secretion after transfection of yak GCs,which will underpin future research on ovarian follicles.
