火麻籽壳多酚不同提取工艺及其抗氧化性 被引量:1

Different Extraction Technologies and in vitro Antioxidant Activity of Polyphenols from Hempseed Threshing Residues

摘要 采用8种不同方法提取火麻籽壳多酚,通过对比不同提取方法的提取率与提取物抗氧化活性,筛选出火麻籽壳多酚的最佳提取方法。采用大孔树脂对最佳提取工艺的多酚粗提物样品进行纯化,确定其纯化条件。结果表明:超声波辅助醇提法多酚提取率最高,多酚提取率为2.34 mg/g。不同提取方法所得多酚提取液对DPPH·和ABTS^(+)·都有清除能力,其中超声波辅助醇提法所得样品在不同浓度水平均表现出较强的自由基清除能力。纯化火麻籽壳多酚的最佳树脂为X-5,其吸附率和解吸率分别为74.5%和44.5%,乙醇解吸液浓度为90%。纯化后多酚对DPPH·和ABTS^(+)·的清除能力与样品浓度之间存在量效关系,其IC_(50)值分别0.38 mg/mL和0.10 mg/mL,纯化后的火麻籽壳多酚具有较高的抗氧化活性。 In this study,eight different extraction methods were used for the extraction of polyphenols from hempseed threshing residues.By comparing the extraction rates and the in vitro antioxidant activities of polyphenols obtained via different extraction methods,the optimal extraction technology was selected.The macroporous resin was used to purify polyphenol crude extract samples,and the purification conditions were determined.Based on the results,ultrasonic alcohol extraction yielded the highest polyphenol extraction rate of 2.34 mg/g.The polyphenol extracts obtained via different extraction methods could scavenge DPPH· and ABTS^(+)·,and the extract obtained via ultrasonic alcohol extraction showed strong scavenging ability at different concentration levels.The most suitable resin for purifying polyphenols from hempseed threshing residues was X-5,with adsorption and resolution rates of 74.5% and 44.5%,respectively.The concentration of the ethanol solution was 90%.The scavenging capacity of purified polyphenols on DPPH·and ABTS^(+)·and the concentration of samples had a dose-response relationship,with IC_(50)values of 0.38 mg/mL and 0.10 mg/mL,respectively.The purified hempseed threshing polyphenols had high antioxidant activity levels.
