摘要
目的:通过对干眼模型兔结膜高迁移率族蛋白B1(HMGB1)的研究,探讨针刺对干眼模型兔结膜炎症的作用机制。方法:选取24只健康新西兰兔,随机分为空白组、模型组、针刺组和西药组4组,各6只。空白组和其余3组每日分别进行皮下0.9%氯化钠溶液及氢溴酸东莨菪碱注射,连续35 d。造模成功后,西药组予氟米龙滴眼液滴双眼,针刺组进行电针治疗(取穴:睛明、攒竹、瞳子髎、丝竹空、太阳,均双侧取穴,留针15 min)。SⅠT检测滤纸条湿润长度和泪膜破裂时间,采用光镜对结膜组织结构进行观察,免疫组化法检测结膜HMGB1蛋白的表达情况,蛋白芯片技术检测白细胞介素(IL)-1β和IL-8的表达情况。结果:与模型组同期比较,针刺组和西药组第35天SⅠT湿润长度和泪膜破裂时间显著增加(P<0.01);模型组结膜组织可见淋巴细胞及中性粒细胞浸润,西药和针刺可减轻结膜淋巴细胞浸润;与模型组比较,针刺组和西药组结膜HMGB1蛋白及IL-1β、IL-8表达均显著降低(P<0.01)。结论:针刺可改善干眼模型兔泪液分泌,减轻结膜炎症,可能与调节结膜HMGB1的表达有关。
Objective:To investigate the mechanism of acupuncture on inflammation of conjunctiva in dry eye model rabbits by studying the high mobility group box 1(HMGB1)signal pathway.Methods:Twenty-four healthy New Zealand rabbits were randomly divided into blank group,model group,acupuncture group and western medicine group,with 6 rabbits in each group.The blank group and the other 3 groups were injected with subcutaneous normal saline and scopolamine hydrobromide(Scop)every day for 35 days.After successful modeling,the medicine group was treated with fluminolone(Flu)eye drops,and acupuncture group was treated with electroacupuncture(EA),acupoints:Taiyang(EX-HN 5),Jingming(BL 1),Cuanzhu(BL 2),Sizhukong(SJ 23),Tongziliao(GB 1).The amount of tear secretion and tear film breaking-up time of experimental animals were detected.The conjunctival tissue structure was observed by light microscope.The expression of conjunctival HMGB1 protein were detected by immunohistochemistry.The expression of IL-1βand IL-8 were detected by protein chip technology.Results:Compared with Scop group,the amount of tear secretion and tear film break-up time in EA group and Flu group were significantly improved(P<0.01);In Scop group,lymphocyte and neutrophil infiltration could be seen.Flu and EA could reduce lymphocyte infiltration;Compared with Scop group,the expression of HMGB1 protein,IL-1βand IL-8 in conjunctiva of EA group and Flu group decreased significantly(P<0.01).Conclusion:Acupuncture can improve tear secretion in dry eye model rabbits,and reduce conjunctival inflammation.The mechanism may be related to the regulation of HMGB1 expression in conjunctiva tissue.
