摘要
以从厂内窖泥中分离、纯化、鉴定获得的克氏梭菌为基础,在实验室人工和制窖泥时,加入克氏梭菌菌液、复合菌液和不添加菌液发酵培养窖泥。以3类窖泥为研究对象,对不同培养时间的3种人工培育的窖泥进行分析检测。结果显示,3个方案的窖泥,各项理化指标随发酵时间呈现出相应并且近似相同的规律。克氏方案有机质、有效磷和氨态氮略高于复合,两者均高于对照,乳酸、正丙醇、正丁醇低于复合和对照;微生物方面,通过培养发现人工窖泥中还存在其他相对比例更高的产己酸菌,比如Ca-proiciproducens、Megasphaera埃氏巨球型菌、Ruminococcus_1瘤球菌1;三组在发酵过程均出现放线菌,添加克氏梭菌窖泥中放线菌丰度高于其他两组。
Clostridium kluyveri strains were isolated and purified from the pit mud of our distillery.Artificial pit mud was made in the laboratory with/without Clostridium kluyveri liquid and compound microbial liquid.The three kinds of pit mud cultured for different time were analyzed.The results showed that the physicochemical indexes of the three kinds of pit mud showed similar change trends with the increase of fermentation time.The contents of organic matter,available phosphorus and ammonia nitrogen in the pit mud made with Clostridium kluyveri liquid were slightly higher than those in the pit mud made with compound microbial liquid,and both were higher than those in the control group.The contents of lactic acid,n-propanol and n-butanol in the pit mud made with Clostridi-um kluyveri liquid were lower than those in the other two kinds of pit mud.Other caproic acid-producing bacteria with higher relative proportions were found in the artificial pit mud,such as Caproiciproducens,Megasphaera and Ruminococcus_1.Actinomycetes was also detected in the fermentation process,and its abundance in the pit mud made with Clostridium kluyveri liquid was higher than that in the other two kinds of pit mud.
