核壳温敏型药物载体负载槲皮素对SiO_(2)诱导的大鼠巨噬细胞极化的抑制作用 被引量:1

摘要 目的:探讨核壳温敏型药物载体负载槲皮素(QC@mSiO_(2)-NIPAM)对SiO_(2)诱导的巨噬细胞极化的作用。方法:大鼠肺泡巨噬细胞NR8383分为空载体组、SiO_(2)+空载体组、SiO_(2)+载体药组,分别给予25μmol/L mSiO_(2)-NIPAM,25μmol/L mSiO_(2)-NIPAM+50 mg/L SiO_(2)混悬液,25μmol/L QC@mSiO_(2)-NIPAM+50 mg/L SiO_(2)混悬液,均孵育24 h。采用Western blot法、免疫细胞化学染色检测M1型、M2型巨噬细胞标记蛋白及MAPK信号通路蛋白的表达。结果:与空载体组比较,SiO_(2)+空载体组M1型巨噬细胞标记蛋白(iNOS、IL-1β、TNF-α),M2型巨噬细胞标记蛋白(精氨酸酶1、IL-10、TGF-β1),MAPK信号通路蛋白(p-ERK1/2、p-JNK、p-P38)表达上调(P<0.05);与SiO_(2)+空载体组比较,SiO_(2)+载体药组M1、M2型巨噬细胞标记蛋白及MAPK信号通路蛋白表达均下降(P<0.05)。结论:QC@mSiO_(2)-NIPAM可能通过调控MAPK信号通路抑制SiO_(2)诱导的NR8383细胞极化。 Aim:To probe the effects of QC@mSiO_(2)-NIPAM on macrophage polarization induced with SiO_(2).Methods:Rat alveolar macrophages were allocated into 3 groups,carrier group(25μmol/L mSiO_(2)-NIPAM),SiO_(2)+carrier group(25μmol/L mSiO_(2)-NIPAM+50 mg/L SiO_(2)),SiO_(2)+carrier drug group(25μmol/L QC@mSiO_(2)-NIPAM+50 mg/L SiO_(2)).After 24 hours,the protein expressions of M1,M2 type macrophage marker proteins and MAPK signaling pathway proteins were detected by Western blot and immunocytochemical staining.Results:Compared with the carrier group,M1 type macrophage marker proteins(iNOS,IL-1β,TNF-α),M2 type macrophage marker proteins(Arginase1,IL-10,TGF-β1),and MAPK signaling pathway proteins(p-ERK1/2,p-JNK and p-P38)showed upregulated expression in the SiO_(2)+carrier group(P<0.05);compared with the SiO_(2)+carrier group,the above indexes were all downregulated in the SiO_(2)+carrier drug group(P<0.05).Conclusion:QC@mSiO_(2)-NIPAM may lead to the inhibition of macrophage polarization by regulating the MAPK signaling pathway.
