新疆南部地区绵羊无形体流行情况调查

Investigation on the prevalence of Anaplasma ovis in southern Xinjiang

摘要 试验旨在确定新疆南部地区绵羊中绵羊无形体基因型种类,共采集240份绵羊血液样本,其中成羊和羔羊各120份样本。样本进行DNA提取,并应用绵羊无形体msp4基因种特异性引物对总DNA进行PCR扩增。测序结果应用DNAstar软件进行核苷酸序列和氨基酸序列比对,并应用MEGA 7.0构建系统进化树。结果显示,绵羊无形体在绵羊中总检出率为22.5%(54/240),其中成年雌性绵羊和羔羊无形体的检出率分别为37.5%(45/120)和7.5%(9/120)。所得序列经比对发现,其同源性为99.7%~100.0%,存在多个核苷酸位点发生改变,并获得3种基因型,分别为基因型Ⅱ型、Ⅲ型、GB3型,各基因型在绵羊中总流行率为5.4%(13/240)、15.0%(36/240)、2.1%(5/240)。研究表明,我国新疆南部地区绵羊中存在3种绵羊无形体基因型,其中基因Ⅲ型是新疆南部地区绵羊中普遍存在的基因型。 The aim of the experiment was to determine the genotypes of anaplasmosis of sheep in southern Xinjiang. A total of 240 sheep blood samples were collected, including 120 samples from adults and 120 samples from lambs. DNA was extracted from the samples, and the total DNA was amplified by PCR using species-specific primers of the sheep anaplasmosis msp4 gene. Nucleotide and amino acid sequences were compared by DNAstar software, and phylogenetic tree was constructed by MEGA 7.0. The results showed that the total detection rate of anaplasmosis in sheep was 22.5%(54/240), and the detection rate of anaplasmosis in adult female sheep and lambs was 37.5%(45/120) and 7.5%(9/120),respectively. The results showed that the sequence homology was 99.7%~100.0%, with multiple nucleotide site changes,and three genotypes were obtained, namely genotypes Ⅱ, Ⅲ, GB3. The overall prevalence of these genotypes in sheep was 5.4%(13/240), 15.0%(36/240), 2.1%(5/240). The study indicates that there were three genotypes of anaplasmosis in sheep in southern Xinjiang, among which genotype Ⅲ was a common genotype in sheep in southern Xinjiang.
