基于生物信息学分析对儿童髓母细胞瘤中差异表达核心基因的筛选及非编码RNA的预测

摘要 目的探讨儿童髓母细胞瘤的治疗靶点。方法采用生物信息学研究方法筛选病例组瘤组织的差异表达基因,对其进行蛋白质互作网络、信号通路、核心基因的鉴定及非编码RNA的预测。结果共获取1307个差异表达基因,其中高表达730个,低表达577个,它们富集在化学突触传递的调节、跨突触信号的调节、微管蛋白结合、微管结合、细胞周期和DNA复制等生物过程和通路上。自蛋白质互作网络中筛选出5个核心基因:CDC20、CDK1、KIF11、ASPM和MAD2L1,最终构建出包含CDK1、KIF11和MAD2L1三个基因、12个miRNAs及60个circRNAs的三个ceRNA网络。结论本研究探索出5个参与儿童髓母细胞瘤发生、发展的关键基因以及可能参与疾病进程的miRNAs、circRNAs等ncRNAs,为儿童髓母细胞瘤的潜在药物治疗靶点提供有力的依据,也为其精准治疗提供了新思路。 Objective To explore the therapeutic target of medulloblastoma in children.Methods Bioinformatic research method was used to screen the differentially expressed genes in tumor tissues of medulloblastoma cases,the protein interaction network,the signaling pathway,and the core genes were identified,and the NcRNA was predicted.Results A total of 1307 differentially expressed genes were obtained,of which 730 showed high expression and 577 showed low expression.They were concentrated in biological processes and pathways of the regulation of chemical synaptic transmission,the regulation of trans-synaptic signals,the tubulin binding,the microtubule binding,the cell cycle and the DNA replication.Five core genes(CDC20,CDK1,KIF11,ASPM and MAD2L1)were screened out from the protein interaction network.Finally,three ceRNA networks were constructed,including the network of CDK1,KIF11 and MAD2L1,the network of 12 miRNAs and the network of 60 circRNAs.Conclusion In this study,five key genes involved in the occurrence and development of pediatric medulloblastoma and ncRNAs such as miRNAs and circRNAs that may participate in the disease process were explored,providing a strong basis for potential drug treatment targets of pediatric medulloblastoma and a new idea for its precise treatment.
