响应面法优化白簕叶总皂苷提取工艺及其大孔树脂静态吸附研究 被引量:2

Optimization of Extraction Process of Total Saponins from Acanthopanax Trifoliatus Leaves by Response Surface Methodology and Study on Static Adsorption of Macroporous Resin

摘要 目的:优化白簕叶中总皂苷的超声提取工艺,并考察不同类型的大孔吸附树脂对总皂苷的静态吸附能力。方法:以白簕叶总皂苷提取率为指标,采用Box-Benhnken响应面法优化乙醇浓度、液料比、超声时间等工艺参数;以大孔树脂对总皂苷的比吸附量和解吸率为评价指标,从14种不同极性的树脂类型中筛选出适用于总皂苷纯化的型号。结果:优化得到最佳提取工艺参数:乙醇浓度为60%,液料比为40 mL/g,提取时间为33 min,此条件下白簕叶总皂苷提取率为5.32%。不同类型的大孔树脂对总皂苷的比吸附量和解吸率存在较大差异,HPD-BJQH、HPD-100和LSA-10型树脂对白簕叶总皂苷的吸附能力和解吸附能力较好。结论:响应面法优化的白簕叶总皂苷超声提取工艺稳定可行,HPD-BJQH、HPD-100和LSA-10型树脂可以作为白簕叶总皂苷大孔树脂纯化工艺中的树脂用料。 Objective:To optimize the ultrasonic extraction process of total saponins from Acanthopanax trifoliatus leaves,and to investigate the static adsorption capacity of different types of macroporous adsorption resins for total saponins.Methods:Taking the extraction rate of total saponins from Acanthopanax trifoliatus leaves as an index,the Box-Benhnken response surface method was used to optimize the process parameters such as ethanol concentration,liquid/solid ratio,ultrasonic time,etc;Based on the specific adsorption capacity and desorption rate of macroporous resin for total saponins,the models suitable for purification of total saponins were selected from 14 resin types with different polarity.Results:The optimum extraction conditions were as follows:ethanol concentration was 60%,extraction time was 33 min,liquid/solid ratio was 40 mL/g.Under this condition,the yield of total saponins from Acanthopanax trifoliatus was 5.32%.The specific adsorption capacity and desorption rate of total saponins on different types of macroporous resins are quite different.HPD-BJQH,HPD-100 and LSA-10 resins have better adsorption capacity and desorption capacity for total saponins from Acanthopanax trifoliatus leaves.Conclusion:The extraction process optimized by response surface methodology is stable,reasonable and feasible.HPD-BJQH,HPD-100 and LSA-10 can be used as resin materials in the purification process of total saponins from Acanthopanax trifoliatus.
