HPLC法测定黄芪桂枝五物汤物质基准6种成分含量 被引量:2

Determination of Six Components in Material Benchmark of Huangqi Guizhi Wuwutang by HPLC

摘要 目的建立同时测定黄芪桂枝五物汤物质基准中芍药内酯苷、芍药苷、毛蕊异黄酮葡萄糖苷、芒柄花苷、肉桂酸、6-姜辣素6种活性成分含量的方法。方法采用Thermo Hypersil GOLD C_(18)色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.2%甲酸溶液,梯度洗脱,检测波长为250 nm(0~35 min,芍药内酯苷、芍药苷、毛蕊异黄酮葡萄糖苷、芒柄花苷)、280 nm(35~50 min,肉桂酸、6-姜辣素),流速为1 mL·min^(-1),柱温为25℃。结果芍药内酯苷、芍药苷、毛蕊异黄酮葡萄糖苷、芒柄花苷、肉桂酸和6-姜辣素分别在29.21~467.4、151.1~2417、1.830~29.28、0.7154~11.45、2.529~40.47、6.157~98.51μg·mL^(-1)线性关系良好,r均≥0.9999,平均加样回收率分别为99.74%、99.71%、101.29%、100.51%、100.40%、98.73%,RSD均小于2.0%。结论该方法简便可靠,灵敏度高,适用于黄芪桂枝五物汤复方制剂研发的质量控制。 OBJECTIVE To establish a method for simultaneously determining the contents of albiflorin,paeoniflorin,calycosin-7-O-β-D-glucoside,ononin,cinnamic acid and 6-gingerol in material benchmark of Huangqi Guizhi Wuwutang.METHODS The quantitative analysis was performed on a Thermo Hypersil GOLD C_(18) column(4.6 mm×250 mm,5μm)with the gradient elution of acetonitrile-0.2%formic acid,the detection wavelengths were 250 nm(0-35 min,albiflorin,paeoniflorin,calycosin-7-O-β-D-glucoside and ononin),280 nm(35-50 min,cinnamic acid and 6-gingerol),the flow rate was 1.0 mL·min^(-1),the column temperature was 25℃.RESULTS The linear ranges of the above 6 components of albiflorin,paeoniflorin,calycosin-7-O-β-D-glucoside,ononin,cinnamic acid and 6-gingerol were 29.21-467.4,151.1-2417,1.830-29.28,0.7154-11.45,2.529-40.47,6.157-98.51μg·mL^(-1),respectively(r≥0.9999).The average recoveries of them were 99.74%,99.71%,101.29%,100.51%,100.40%,98.73%,respectively(RSD<2.0%).CONCLUSION The established method is reliable,which has the characteristics of easy maneuverability and high sensitivity.The method can be used for the quality control of Huangqi Guizhi Wuwutang preparations.
