摘要
该实验的主要目的在于探索开发科学控制组织培养中培养基杂菌污染的新配方。新型的配方研发实验需要对其原有的液体进行一定的还原处理工作,然后再将处理过后得到的新型粒子加入到内含植物组织的载体培养基中,进而得到一种对杂菌性质具备抗体能力的复合培养基。需要利用现有的科学技术手段,对培养基内的污染因子加以管理和控制,从而能够形成不用管控植物组织就可以自行成长的开放式培养模式。新配方实验结果表明,伴随硝酸银的溶液浓度变大,纳米银的粒子粒径尺寸随之变大,粒径的具体分布范围明显变宽。应用于控制组织培养中培养基杂菌污染中进行抗菌的测试结果说明,纳米银粒子复合的B5培养基明显具有抗杂菌污染的性能,组织培养材料被污染的风险被明显降低,该配方的制备方法对已经污染的组织培养材料的挽救和大田种质材料实验室保存有重要价值。
The main purpose of this experiment is to explore and develop a new formula to scientifically control the contamination of culture medium in tissue culture. The new formula research and development experiment requires a certain reduction treatment on its original liquid, then the new particles obtained after treatment are added to the carrier medium containing plant tissue, and a compound culture medium with antibody ability to properties of bacteria is obtained. It is necessary to use existing scientific and technological means to manage and control the pollution factors in the culture medium, so as to achieve an open culture mode which allows the plant to grow without controlling its tissues. The experimental results of the new formula show that with the increase of the solution concentration of silver nitrate, the particle size of silver nanoparticles increases, and the specific distribution range of particle size becomes significantly widener. The test results applied to the control of miscellaneous bacteria contamination in the culture medium in tissue culture show that the B5 medium compounded by nano-silver particles has obvious resistance to miscellaneous bacteria contamination, and the risk of contamination of tissue culture materials is significantly reduced. The preparation method of this formula is of great value to the rescue of contaminated tissue culture materials and the laboratory preservation of field germplasm materials.
