摘要
肿瘤是威胁人类生存的主要疾病之一,近年来,尽管已针对肿瘤发病机制进行了深入的研究,并持续开发并应用不同种类的抗肿瘤药物,但其致死率仍居高不下。肿瘤发生发展主要受原癌基因活化、抑癌基因失调、肿瘤生长过程中的代谢环境改变及肿瘤免疫逃逸等多种因素调节。自噬是一种细胞内成分降解和循环机制,其可通过自噬体的双层膜结构包裹并降解细胞内容物,释放循环营养物质,帮助维持细胞稳态及正常生理过程。越来越多的研究表明,肿瘤微环境中自噬与肿瘤发生发展息息相关。文章阐述了自噬与肿瘤发生发展的相关性,为肿瘤研究及肿瘤临床治疗提供有力的理论基础。
Malignant tumor remains one of the most life-threatening diseases.Despite intensive research aimed at pathogenesis and drug treatment has been conducted for decades,the patients who underwent melanoma remain the high mortality.Neoplastic progression is mainly induced by the dysregulation of the proto-oncogenes,tumor suppressor genes,the altered metabolic environment,immune escape,and etc.As an intracellular recycling and degradation mechanism,autophagy can maintain cellular homeostasis and normal physiological processes through autophagosome.The double-membrane wraps can degrade and release the contents and further facilitate to circulate nutrients.There is the growing evidence that autophagy in the tumor microenvironment is related to tumor development.This review aims to clarify the correlation between autophagy and tumor development and provide the strong theories for tumor research and clinical treatment.
