摘要
目的探讨p57Kip2基因与妊娠期糖尿病(GDM)之间的相关性。方法本研究为病例对照研究,选取2017年3月至2018年12月济宁医学院附属医院产科收治的60名孕妇,年龄(29.95±4.66)岁,年龄范围为18~38岁。依据是否患有GDM分为GDM组与健康组,GDM孕妇30例,健康孕妇30名。分娩后取胎盘样本并制作石蜡组织切片,通过免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结法(SP)法对p57Kip2蛋白在胎盘组织中的表达含量及细胞定位进行检测;通过逆转录PCR法(RT-PCR)检测GDM患者胎盘组织及健康孕妇胎盘组织中p57Kip2 mRNA的表达水平;分析p57Kip2与孕妇空腹血糖及新生儿体质量的相关性。结果免疫组化染色结果显示,p57Kip2蛋白主要表达于胎盘组织的细胞滋养细胞的细胞核内,在合体滋养细胞中表达较少。GDM孕妇胎盘组织p57Kip2蛋白的表达水平[(0.24±0.15)nmol/L]显著高于健康孕妇胎盘组织[(0.16±0.09)nmol/L],差异有统计学意义(t=-2.54,P=0.013)。GDM孕妇胎盘组织p57Kip2 mRNA的表达水平显著高于健康孕妇胎盘组织,差异有统计学意义(P=0.012)。GDM组空腹血糖[(5.10±0.43)mmol/L]的含量高于健康组[(4.33±0.39)mmol/L],差异有统计学意义(P<0.05),两组纳入者新生儿体质量比较,差异无统计学意义(P>0.05)。结论p57Kip2蛋白和p57Kip2 mRNA在GDM患者胎盘组织中的表达水平高于健康孕妇的胎盘组织,提示p57Kip2基因可能参与了GDM的发病过程。
Objective To investigate the expression level of p57Kip2 gene in placenta tissues of normal pregnant women and gestational diabetes mellitus(GDM)patients,we will explore the correlation between p57Kip2 gene and GDM.These results will provide a basis for p57Kip2 as an early monitoring indicator of GDM and a new therapeutic drug target.Methods This study was a case-control study.A total of 60 pregnant women,aged(29.95±4.66)years old,ranging from 18 to 38 years old,who received the obstetrics department of Affiliated Hospital of Jining Medical College from March 2017 to December 2018.They were divided into GDM group and healthy group according to whether they had GDM or not,there were 30 GDM pregnant women and 30 healthy pregnant women.After delivery,placenta samples were taken and paraffin sections were prepared.The expression and localization of p57Kip2 protein in placenta tissues were detected by immunohistochemical streptavidin-perosidase(SP)method.The expression level of p57Kip2 mRNA in the placental tissues of GDM patients and healthy pregnant women was detected by reverse transcription polymerase chain reaction(RT-PCR).To analyze the correlation between p57Kip2 and fasting blood glucose in pregnant women and neonatal weight.Results Immunohistochemical staining showed that p57Kip2 protein was mainly expressed in the nucleus of cytotrophoblast cells of placental tissue,but was less expressed in syncytiotrophoblast cells.The expression level of p57Kip2 protein in placental tissue of GDM pregnant women[(0.24±0.15)nmol/L]was significantly higher than that of healthy pregnant women[(0.16±0.09)nmol/L],and the difference was statistically significant(t=-2.54,P=0.013).The expression level of p57Kip2 mRNA in the placental tissues of GDM pregnant women was significantly higher than that of healthy pregnant women,and the difference was statistically significant(P=0.012).The content of fasting blood glucose[(5.10±0.43)mmol/L]in GDM group was higher than that in healthy group[(4.33±0.39)mmol/L],the difference was statistically significant(P<0.05),and there was no statistically significant difference in neonatal weight between the two groups(P>0.05).Conclusions The expression levels of p57Kip2 protein and mRNA in the placental tissues of patients with GDM were higher than those of healthy pregnant women,which suggests that p57Kip2 gene may be involved in the pathogenesis of GDM.
