不同来源猫爪草多糖质量分数测定及其抗氧化活性研究

摘要 以猫爪草为材料,研究不同提取时间、液料比、提取温度对猫爪草多糖质量分数的影响。在单因素试验的基础上,以猫爪草的多糖质量分数作为响应值,运用响应面法对猫爪草多糖提取条件进行优化。采用苯酚-硫酸法测定10个不同来源猫爪草药材中多糖的质量分数,并利用猫爪草多糖对DPPH的清除活性评价其抗氧化作用。结果表明:超声提取猫爪草多糖的最佳条件是提取时间41 min,液料比15∶1 mL·g^(-1),提取温度84.6℃。不同来源猫爪草多糖的质量分数和抗氧化活性差异明显,其中河南信阳淮滨多糖质量分数最高,为72.72 mg·g^(-1),IC50为2.6 mg·mL^(-1),且猫爪草多糖对DPPH的清除能力与多糖质量分数呈正相关性。 Taking R.Ternati as material,the influence of different extraction time,liquid-to-material ratio and extraction temperature on the mass fraction of R.Ternati polysaccharide was studied.Based on the single factor test,the polysaccharide mass fraction of R.Ternati polysaccharides was used as the response value,and the response surface method was used to optimize the extraction conditions of R.Ternatipolysaccharides.The phenol-sulfuric acid method was used to determine the mass fraction of polysaccharides in R.Ternati from different sources,and the antioxidant effect was evaluated by DPPH scavenging activity.The results showed that the optimal conditions for ultrasonic extraction of polysaccharide were as follows:extraction time was 41 min,liquid-solid ratio was 15∶1 mL·g^(-1),extraction temperature was 84.6℃.There were significant differences in polysaccharide mass fractions and antioxidant activity of the R.Ternati from different producing areas.The polysaccharide mass fraction of Huaibin in Xinyang,Henan was the highest,72.72 mg·g^(-1),with IC502.6 mg·mL^(-1),and the scavenging ability of R.Ternati polysaccharide on DPPH was positively correlated with the polysaccharide mass fraction.
