马疱疹病毒1型gD囊膜蛋白互作蛋白的鉴定分析

摘要 旨在筛选并分析与马疱疹病毒1型(EHV-1)gD囊膜蛋白相互作用的宿主蛋白。用免疫共沉淀和LCMS/MS质谱技术鉴定出与EHV-1 gD囊膜蛋白互作的宿主蛋白,并进行生物信息学分析。免疫共沉淀联合质谱分析鉴定到229个与EHV-1 gD囊膜蛋白互作的宿主蛋白,对蛋白功能分析发现大多数蛋白与形成细胞骨架有关,细胞骨架不仅能支撑细胞形态,还参与物质运输,结果提示有部分蛋白参与囊泡运输并在其中发挥重要作用,还存在与囊泡形成直接相关的蛋白——网络蛋白重链,为探究EHV-1二次囊膜化的形成过程和研究gD囊膜蛋白与囊泡间的关系奠定了基础。 The aim of this study was to screen and analyze the proteins that interact with EHV-1 gD envelope protein.We used co-immunoprecipitation and liquid chromatography-tandem mass spectrometry to screen out the host proteins interacting with the gD envelope protein of the EHV-1,and analyzed by bioinformatics.The results showed that immunoprecipitation combined with mass spectrometry identified 229 host proteins that interacted with EHV-1 gD envelope protein,and further analysis of protein functions in this study revealed that most of the proteins are related to the formation of cytoskeleton,which not only supports cell morphology but also participates in material transport.The analysis results also suggest that some proteins are involved in vesicle transport and play an important role in it,and there is another protein directly related to vesicle formation,Clathrin heavy chain.The paper establishes the foundation for studying the formation process of secondary vesiculation of EHV-1 and provides a direction for studying the relationship between gD envelope proteins and vesicles.
