耧斗菜WRKY转录因子家族生物信息学分析及抗盐基因筛选 被引量:1

摘要 为挖掘耧斗菜属植物WRKY的抗盐基因奠定理论基础,通过转录组数据分析,鉴定WRKY转录因子,并对家族成员进行生物信息学分析。结合表达模式对其功能进行分析,结果表明:从欧耧斗菜中鉴定出35个WRKY转录因子,根据WRKY七肽的数量和锌指结构特点可分为3亚族,每组成员分别有8、24、3个;其编码蛋白质氨基酸数为173~797个,分子量为19902.13~87973.25,均为亲水性蛋白,其中有1个稳定蛋白;亚细胞定位结果均在细胞核中。结合系统进化树,从14个差异显著表达的AvWRKY基因中,筛选出与已知抗盐的WRKY基因亲缘关系近的4个基因(AvWRKY3、AvWRKY7、AvWRKY16、AvWRKY34),作为在耧斗菜响应盐胁迫中发挥重要作用的候选基因。 To provide theoretical basis for mining the salt resistance gene of WRKY in Aquilegia,we identified WRKY transcription factors through analysis of transcriptome data,and analyzed family members by bioinformatics.Combined with expression patterns,we analyzed its function.The results showed that 35 WRKY transcription factors were identified from A.vulgaris,which could be divided into 3 subfamilies according to the number of WRKY motif and zinc finger structure.Each group has 8,24 and 3 members,respectively.The amino acids of deduced protein ranged from 522 to 3610,and the molecular weight was 42963.9-296709.62.They were all predicted to be hydrophobic proteins,including 12 stabilizing proteins.Besides,subcellular localizations were all in the nucleus.Combined with the phylogenetic tree,we screened out four genes(AvWRKY3,AvWRKY7,AvWRKY16 and AvWRKY34)from 14 AvWRKY with significant differential expression,which were closely related to the known salt resistance WRKY genes,and to be as candidate genes that play an important role in the response to salt stress in Aquilegia.
