MCM6促进食管癌细胞增殖进而导致预后不良 被引量:1

MCM6 Contributes to the Proliferation of Esophageal Carcinoma Cells and Predicts Poor Prognosis

摘要 食管癌是一种常见的致死率高的恶性肿瘤.为确定食管癌发生的潜在机制,在生物信息学分析的基础上筛选关键基因.染色体维持6复合体(Chromosome maintenance 6 complex,MCM6)在多种恶性肿瘤中上调,但是,它在食管癌中的作用仍然不清楚.利用GEPIA数据库,研究MCM6基因在正常食管组织和癌性食管组织中的表达变化.结果表明,食管癌中MCM6基因有明显的高表达水平.利用免疫组化方法检测MCM6蛋白在正常食管组织及恶性肿瘤组织中的表达,发现食管癌中MCM6蛋白有明显的高表达水平.用卡方检验分析临床病理特征与蛋白表达的关系,发现本院78例患者MCM6表达与食管癌肿瘤大小有显著相关性.通过集落形成试验,确定MCM6能促进食管癌细胞的增殖.肿瘤生长试验证实了MCM6在小鼠食管癌进展中的促进作用.通过数据库和免疫组化检测MCM6与PCNA的相关性,发现MCM6与PNCA基因和蛋白呈正相关,进而推测MCM6可能通过影响PNCA的表达促进食管癌的增殖.综上所述,MCM6可能成为治疗食管癌的新靶点. Esophageal carcinoma(ESCC) is a common type of fatal cancer with high mortality. The purpose is to identify key genes and determine the potential mechanisms involved in the development of esophageal carcinoma based on bioinformatics analysis. Chromosome maintenance 6 complex(MCM6) has been identified to be up-regulated in a variety of malignancies. However, its effects on esophageal carcinoma remain unclear.Using the GEPIA database, it was investigated how MCM6 expression varied between healthy and cancerous esophagus tissues. And the result that ESCC with high MCM6 gene expression was found. Then immunohistochemistry(IHC) assay was performed to detect MCM6 expression in both healthy and malignant esophageal tissues, and the result showed high MCM6 protein expression in ESCC. The χ2 analysis to explore the correlation between clinic-pathological features and protein expression and the tumor size of esophageal cancer was substantially associated with MCM6 expression in 78 patients in our hospital. By performing a colony formation assay, the promoting effect of MCM6 on the proliferation of esophageal cancer cells was identified.Tumor growth assay confirmed the promoting role of MCM6 in the progression of esophageal cancer in vivo.The correlation between MCM6 and PCNA was detected by database and immunohistochemistry, and we found that MCM6 was positively correlated with PCNA expression. Further, it was speculated that MCM6 might promote the proliferation of esophageal cancer cells by mediating PNCA expression. In conclusion, MCM6 might become a new target for the treatment of esophageal carcinoma.
