基于LC-MS/MS技术的武平紫灵芝子实体化学成分分析 被引量:1

摘要 采用药典规定方法、测定试剂盒及LC-MS/MS方法对武平紫灵芝各类化学成分的组成及含量进行分析,结果表明:与同样来源赤灵芝相比,紫灵芝的灵芝多糖、总黄酮和总氨基酸含量更高,三萜酸含量较低。两者的化学组成成分相似,其主要三萜为灵芝酸B和灵芝酸C,灵芝酸A含量显著低于赤灵芝(P<0.05);多糖主要由葡萄糖、半乳糖、甘露糖、阿拉伯糖和岩藻糖组成,其中甘露糖、阿拉伯糖含量显著高于赤灵芝(P<0.05)。黄酮类中含有柚皮素、黄豆黄苷、山奈酚、表儿茶素、甘草素等成分,其中柚皮素和表儿茶素L含量显著高于赤灵芝(P<0.05)。氨基酸含量组成无显著差异(P>0.05)。LC-MS/MS分析表明,与同样来源赤灵芝相比,紫灵芝三萜类含量增加最多的是2,2'-脱水尿苷和刺囊酸,降低最多的是灵芝酸A;糖类增加最多的是阿拉伯糖,降低最多的是岩藻糖;黄酮类增加最多的是柚皮苷,降低最多的是山奈酚;氨基酸增加最多的是丙氨酸,未有差异2倍以上含量降低的氨基酸。 In this paper,the composition and content of various chemical components of Ganoderma sinense from Wuping were analyzed by the methods specified in the Pharmacopoeia,the determination kit and LC-MS/MS methods.The results showed that the content of polysaccharide,total flavone and total amino acid of Ganoderma sinense was higher than that of Ganoderma lucidum from the same source,while the content of triterpene acid was lower.The chemical compositions of the two samples were similar.The main triterpenoids of Ganoderma sinense were Ganoderma lucidum acid B and Ganoderma lucidum acid C.The content of Ganoderma lucidum acid A was significantly lower than that of Ganoderma lucidum(P<0.05).Polysaccharides of Ganoderma sinense were mainly composed of Glucose,Galactose,Mannose,Arabinose and Fucose,and the contents of mannose and Arabinose were significantly higher than those of Ganoderma lucidum(P<0.05).The flavonoids of Ganoderma sinense contain Naringin,Daidzein,Kaempferol,Epicatechin,Licorice and other components,among which the contents of Naringin and Epicatechin L were significantly higher than those of Ganoderma lucidum(P<0.05).There was no significant difference in amino acid content between the two samples P>0.05).Compared with Ganoderma lucidum from the same source,LC-MS/MS analysis showed that the content of triterpenoids in Ganoderma sinense increased most was 2,2'-dehydrated uridine and Echinocandin,and decreased most was Ganoderma lucidum acid A.The largest increase in glucide was Arabinose,and the most decreasing was Fucose.The most increased flavonoids were Naringin,and the most decreasing was Kaempferol.And the largest increase in amino acids was Alanine,and there was no difference in amino acids with a decrease of more than 2 times.
